摘  要:目的探讨过氧化物酶体增殖物激活受体γ(PPARγ)激动剂罗格列酮对脑缺血再灌注损伤的保护作用。方法采用线栓法制作小鼠大脑中动脉栓塞模型(MCAO)。将健康成年昆明小鼠随机分为缺血对照组、溶剂对照组(二甲基亚枫)、罗格列酮组。再灌注24小时后观察小鼠神经功能评分,2,3,5三苯基四氮唑(TTC)染色法测量脑梗死体积,干湿重法测量缺血侧脑含水量,常规HE染色观察病理变化。结果罗格列酮组小鼠神经功能评分明显低于缺血对照组、溶剂对照组(P均<0.01),脑梗死体积与其它两组相比分别降低了41%和39%(P均<0.01),脑水含量也明显降低(P均<0.05),脑组织病理学显著改善。结论罗格列酮对小鼠脑缺血再灌注损伤具有明显的保护作用。Objective To study the protective effects of Rosiglitazone,a synthetic ligand of peroxisome proliferator activated receptor γ on focal cerebral ischemia-reperfusion injury in mice. Methods The middle cerebral artery(MCA) was occluded with thread according to modified Zealonga's method. The mice were randomaly divided into three groups: ischemia model group, solvent(DSMO) group,Rosiglitazone pretreatment group. The change of neurological scorns,infarct volume and brain water content were measured at 24 hours after reperfusion. At the same time, pathological change was observed. Results Rosiglitazone pretreatment group can decrease neurological scores ( P 〈 0.01), relieve cerebral infarction volume(P 〈 0.01), subtract brainedma ( P 〈 0.05 ), ameliorate the pathology. Conclusions All the results display Rosiglitazone has the protective effect on focal cerebral ischemia reperfusion injury in mice.
