摘 要:为了测定具有溶磷能力的相思根瘤菌的16S rDNA序列。在TY培养基上将菌株培养至对数生长期,以提取的总DNA为模板,采用双向引物进行PCR扩增,对获得的目的产物进行纯化和测序。采用PHYLIP软件中DNADIST程序分析各菌株间的相似性,采用Clustal X和Treeconw软件获得菌株的系统发育树状图。系统发育学分析表明:G7-3、G31-1-5、G31-1-10位于根瘤菌属(Rhizobium)分支中;F7-2-11位于慢生根瘤菌属(Bradyrhizobium)分支;DH001、LH001、LL007酸、Y木1F、3-2-6位于Mesorhizobium分支中;H1-1-1属于类芽孢杆菌(Paenibacillussp.)。该研究初步确定了具有溶磷能力的相思根瘤菌菌株的发育学地位。The objective of the research was to determine the 16S rDNA sequence of Acacia rhizobium with the ability of phosphorus-solubilizing. The strain was cultured in the TY culture medium until logarithm growth stage, With extracted total DNA as ten,plate, bidirectional primers were used in PCR amplification and the obtained target products were purified and sequenced. The comparabilities among the strains were analyzed with DNADIST program in PHYLIP software and phylogenetic dendrogram was obtained with Clustal X and Treeconw softwares.Phylogenetic analysis showed that G7-3,G31-1-5 and G31-1-10 were in the branch of Rhizobium, F7-2-11 was in the branch of Bradyrhizobium, DH001 ,LH001 ,LI.1307 acid, Y wood 1 and F3-2-6 were in the branch of Mesorhizobium and H1-1-1 was in the branch of Paenibacillus sp.The research confimned the auxology position of the acacia rhizobium with the ability of phosphoms-solubilizing.
