摘  要:采用实时荧光定量PCR技术检测仔猪血液淋巴细胞HSP70 mRNA的转录量,探讨不同冷应激强度对淋巴细胞HSP70 mRNA转录量的影响.结果表明,仔猪在-10^-4℃冷暴露0.5h,淋巴细胞HSP70 mRNA的转录量迅速升高(P<0.05),并一直保持高水平至6h(P<0.05),随后下降,冷暴露结束后24h恢复正常.在4~10℃冷暴露前后,淋巴细胞HSP70 mRNA转录量无明显变化(P>0.05).结果提示,冷暴露温度越低,淋巴细胞中HSP70 mRNA转录越迅速,但冷暴露结束进入常温后HSP70 mRNA转录水平再次升高的时间延后;HSP70 mRNA转录呈现规律性变化,可以考虑把HSP70 mRNA优先作为评估不同冷应激的客观指标.HSP70 mRNA transcription in lymphocyte of piglets before and after cold stress was measured by RFQ-PCR to explore the effect of cold stress with different intensities on the transcription level in piglet lymphocyte. The result indicated that the transcription level increased sharply (P 〈 0.05 ) after piglets were exposed to- 10~-4℃ for 0.5 h. The increased level maintained highest for 6 h ( P 〈 0.05 ) , then declined and recovered to normal at 24 h after cold exposure was terminated. When the animals were exposed to temperature between 4 ℃ and 10 ℃ , there was no significant change in lymphocyte HSP70 mRNA transcription level before and after cold stress (P 〉 0.05 ). The results showed that the lower the cold stress temperature, the quicker the lymphocyte HSP70 mRNA transcription. However, when the cold exposure was completed and the animals were again exposed to normal temperature, there was a delay in rebound time for another lymphocyte HSP70 mRNA transcription. The HSP70 mRNA transcription showed a regular pattern, and it could be used as an objective parameter to evaluate cold stress conditions. Fig 1, Tab 1, Ref 14.
