卵巢原发癌和转移癌中Survivin的表达与Caspase-3、血管内皮生长因子的相关性研究

摘　　要：目的探讨凋亡抑基因Survivin mRNA在卵巢原发癌和转移癌中的表达及其与凋亡终末效应器(Caspase-3mRNA)、血管内皮生长因子(VEGF)表达的相关性。方法应用原位杂交技术检测Survivin mRNA、Caspase-3mRNA、免疫组化S-P法检测VEGF蛋白在卵巢正常组织、良恶性肿瘤中的表达。结果Survivin、Caspase-3、VEGF在卵巢正常组织、良性肿瘤与卵巢恶性肿瘤(原发癌和转移癌)中的表达比率均有统计学意义P<0.01。3种基因表达在卵巢原发癌中与临床分期有关,VEGF表达与淋巴结转移有关。统计学分析显示:Survivin与Caspase-3的表达呈负相关关系P<0.01,r=-0.434,Survivin与VEGF的表达呈正相关关系P<0.05,r=0.317。结论Survivin、VEGF在卵巢原发癌和转移癌中高表达,与临床分期关系密切,可能参与卵巢恶性肿瘤的发生发展和转移,是重要的预后因子。Caspase-3在卵巢恶性肿瘤中低表达,与Survivin的负相关关系提示,Survivin在肿瘤凋亡过程中承担其重要作用。VEGF的过表达对探讨卵巢恶性肿瘤血管形成机制及对肿瘤血管生成的治疗具有重要价值。[ Objective ] To discuss the expression of Survivin mRNA and its correlation with expression of Caspase-3, vascular endo thelial cell growth factor （VEGF） in primary and metastatic ovarian carcinoma. [Methods] The expression of Suvivin and Caspase-3 was detected by in situ hybridization and the expression of VEGF was detected by immunohistochemical assay in normal ovary, ovarian benign and malignant tumor and metastatic ovarian carcinoma. [Results] The positive expression of Survivin, Caspase-3 and VEGF in ovarian carcinoma （primary and metastatic） were statistically significant with that in normal and benign tumor of ovary （P 〈0.01）. The positive expression of them was associated with clinical stage in primary ovarian carcinoma. The expression of VEGF as associated with lymph node metastasis. The analysis showed： Survivin was closed associated negatively with Caspase-3 （r =0.434, P 〈0.01）. Survivin was closely associated positively with VEGF （r =0.317）. [Conclusion] Suvivin and VEGF are highly expressed in primary and metastatic ovarian carcinoma, and may play an important role in the onset and progression of ovarian malignant tumor and be important prognosis factors. The closed associated negatively with Caspase-3 suggests survivin may play an important role in tumor apoptosis. THE over expression VEGF may have important value to approach the mechanism of ovarian carcinoma angiogenesis and treatment of tumor angiogenesis.
