摘 要:为提高根癌农杆菌介导的甘蓝型油菜遗传转化效率,以甘蓝型油菜"湘油15"下胚轴为外植体,以MS为基本培养基,研究了共培养基中α-NAA和6-BA浓度对根癌农杆菌介导法转化甘蓝型油菜过程中下胚轴GUS基因瞬间表达及转化效率的影响。结果表明,共培养基对GUS基因的瞬间表达和稳定表达具有重要影响,但共GUS基因的瞬间表达率和转化效率之间没有明显的线性关系。 In order to improve the transformed efficiency mediated by Agrobacterium tumefaciens,the hypocotyls of a Brassica napus variety Xiangyou 15 were used as the explants and MS was used as the basic medium in the experiment.The effect of the concentration of 6-BA and α-NAA in the co-culture medium on the transient expression frequency and transformed efficiency of transformed GUS gene was investigated.The results showed that the concentration of the phytohormones had significant effect on transient expression frequency and transformed efficiency of transformed GUS gene.But the relation between the transient expression frequency and transformed efficiency was not significant.
