摘 要:目的建立一种较为简单的测定葱白提取物中甾体总皂苷含量的方法。方法以薯蓣皂苷元作为对照品,通过正交实验对香草醛比色法实验条件进行优化,确定比色条件为挥干溶剂后加0.5%香草醛-冰醋酸0.2 mL和高氯酸1.0 mL,于80℃水浴30 m in。结果测得葱白提取物中总皂苷含量为11.50%,平均加样回收率为101.30%,RSD为2.08%(n=6)。结论采用香草醛显色法测定葱白提取物中总皂苷含量操作简便,结果可靠。Objective To establish a simple method of determination on total saponin in Fistular onion bulb by eolorimetrie method. Methods Using diosgenin as a standard substance , the reaction condition of vanillin eolorimetrie method was optimized through orthogonal experiment. The reaction condition was to heat samples heat for 30 min at water bath of 80 ℃ after adding 0.5% vanillin- glacial acetic acid 0, 2 mL and glacial acetic acid 1.0 mL. Results The concentration of total saponin in fistular onion bulb extract was 11.50% ,and the average recovery was 101.30% with relative standard deviation of 2.08% ( n = 6 ). Conclusion This colorimetry method is simple and reliable to measure the concentration of total saponin in fistular onion bulb.
