去甲斑蝥素对裸鼠胰腺癌种植瘤生长的影响

摘　　要：目的探讨去甲斑蝥素(NCTD)对人胰腺癌(AsPC-1细胞)种植瘤增殖和凋亡的影响。方法建立人胰腺癌裸鼠种植瘤模型,实验分为对照组、5-FU组、NCTD组和NCTD+5-FU组。测定种植瘤大小、生长曲线和抑瘤率,应用流式细胞术、光学和电子显微镜观察种植瘤细胞的细胞周期、凋亡率和形态学改变。结果NCTD可使裸鼠种植瘤明显缩小[(2.22±0.18)vs.(3.25±0.18)cm3,P<0.01],抑瘤率增高(31.69%vs.0.0%,P<0.05);种植瘤细胞周期中G2+M期细胞明显增多[(51.24±1.92)%vs.(23.31±1.81)%,P<0.01],凋亡率上升[(0.23±0.05)%vs.(4.21±0.52)%,P<0.01];与5-FU合用时上述作用更明显。NCTD作用后的种植瘤细胞经光镜检查见胞核固缩,核碎裂等现象;电镜可见典型的凋亡细胞及凋亡小体。结论NCTD可明显抑制裸鼠胰腺癌种植瘤的增殖和生长,若与5-FU合用呈协同效应,可加强其抗癌作用。Objective To explore the effect of norcantharidin （NCTD） on proliferation and apoptosis of jimplanted human pancreatic cancer in nude mice. Methods AsPC-1 cells of human pancreatic cancer were implanted subcutaneously into nude mice. Mice were randomly divided into control,5-FU,NCTD and NCTD＋5-FU treatment groups. Tumor size, growth curve and inhibitory rate were evaluated. Cell cycle and apoptosis were measured. Morphological changes of tumor cells were observed. Results Tumor volume was diminished [（2.22±0. 18） vs. （3.25±0. 18） cm^3 ,P〈 0. 01], tumor inhibitory rate was increased （31.69% vs. 0. 0%, P〈 0.05） . The increase in the percentage of the G2 ＋M phase cells [ （51.24 ± 1.92） vs （23. 31 ± 1.81 ）%, P〈 0.01 ）] and cell apoptosis rate [（0. 23±0.05） % vs. （4.21±0. 52） % ,P〈0.01] in NCTD group was greater than that in control group. Compared with other groups, the difference in tumor volume, tumor inhibitory rate, percentage of the G2 ＋M phase cells and cell apoptosis rate in NCTD＋5-FU group was statistically significant（P〈0. 01）, with increased nuclear shrinkage, karyorrhexis and typical apoptosis. Conclusion NCTD inhibits the growth of implanted tumor of human pancreatic cancer in nude mice. The inhibitory effect could be intensified when combined with 5-FU.
