摘  要:目的:探讨辛伐他汀对兔心肌梗死后心功能及梗死周边血管新生的影响。方法:21只中国雌性白兔,随机分为:Ⅰ组(假手术组)、Ⅱ组(实验时照组)、Ⅲ组(辛伐他汀干预组)。Ⅰ组只打开胸腔及心包不造成心肌梗死,Ⅱ组和Ⅲ组为心肌梗死动物模型。Ⅰ、Ⅱ组给予安慰剂,Ⅲ组给予辛伐他汀5 mg·kg^(-1)·d^(-1)。所有兔在饲养8周后处死。测量指标:①术前及梗死后8周的血脂;②梗死边缘区微血管密度;③术后第1、2、4、8周用超声心动图测定左室射血分数(LVEF)。结果:①Ⅲ组的血脂水平在用药八周后明显下降(P<0.05),Ⅰ、Ⅱ组血脂水平前后无明显改变(P>0.05)。②在梗死边缘区Ⅲ组的微血管数目明显多于Ⅱ组。③整个实验过程中Ⅰ组兔的心功能无变化。Ⅱ、Ⅲ组兔的心功能在术后第1周末相近但明显低于Ⅰ组。第2周末Ⅲ组兔的心功能稍好于Ⅱ组,但未达到统计学差异。第4周末,Ⅲ组兔的心ⅱ肌梗死后心功能不全的发生发展。结论:使用辛伐他汀可以增加兔心肌梗死后梗死周边微血管数目。Objective: Evaluating the effects of simvastatin on cardiac function and angiogenesis in rabbits after experimental myocardial infarction. Methods:21 Chinese female rabbits, 2.0 to 2.5 kg in body weight were divided into three groups:groupi was the sham-operated group, which thoracic cavity and pericardial cavity were opened. Group Ⅱ was myocardiol infarction (MI) group, and group Ⅲ was MI + simvastatin-treated group(given simvastatin 5 mg·kg^-1·d^-1). The MI model was created by liquid nitrogen directly freezing myocardium. After eight weeks of treatment the animals were sacrificed. Serum cholesterol levels were at baseline and 8 weeks after operation were measu. The count of microvessels near the margin of the infarcted myocardium which were evaluated by immunohistochemical exams 8 weeks after operation. Left ventricular end-diastolic diameter (LVDD) left ventricular end-systolic diameter(LVSD ) which were measured by echocardiography at 1 week, 2 weeks, 4 weeks, 8 weeks respectively after operation. Results : (1)The serum cholesterol levels in group m were significantly decreased after 8 weeks treatment, but in group Ⅰ and group Ⅱ there was no changes, while microvessel counting in group Ⅲ was significantly higher than group Ⅱ. (2)The cardiac function in group Ⅰhad no changes during all study period. At first week,group Ⅱ and group Ⅲ have similar decreased cardiac function as compared with group Ⅱ. At the end of second week, the cardiac function of group m showed better tendency than that of group Ⅱ. At the end of fourth week, there was a significant difference between group Ⅱ and group Ⅲ. At eighth week, the difference become small,but the cardiac function of Simvastatin-treated group was better than that in control group. Conclusion: Short-term simvastatin therapy can retard the progression of cardiac dysfunction in rabbit MI model. Simvastatin therapy can induce effective angiogenesis near the margin of the infarcted myocardium in rabbit MI mod
