摘 要:目的:检测脂肪源Flk1CD31CD34细胞是否具有血液血管干细胞的特性。方法:将脂肪源Flk1CD31-CD34-细胞分别于血管内皮及造血细胞诱导培养体系中培养,采用RT-PCR、免疫组织化学方法检测分析其生物学特性。结果:脂肪源Flk1+CD31-CD34-细胞在内皮细胞诱导体系中可分化为CD31+/vWF+细胞,在基底膜胶中可形成血管样结构;在造血诱导培养体系中可定向分化为表达GATA-1、GATA-2、β及γ珠蛋白的细胞,在甲基纤维素造血培养基中,这类造血细胞可定向分化为红系集落单位。结论:脂肪源Flk1+CD31-CD34-细胞在体外可同时向血管内皮细胞和造血细胞分化,具有血液血管干细胞的特性。Aim : To investigate whether Flk1^+CD31^-CD34^- cells isolated from adult adipose tissue have characteristics of hemangioblasts. Methods:Flk1^+CD31^-CD34^- cells were characterized on extracellular matrix (ECM) gel or semisolid media supplemented with endothelial or hematopoietic growth factors, respectively. Immunochemistry staining and RT-PCR were performed for cell characterization. Results: After being cultured on endothelial condition, the induced cells were positive for CD31 and vWF. On ECM gel, Flk1^+CD31^-CD34^- cells could grow into vascular structure. Additionally, Flk1^+CD31^-CD34^- cells could differentiate into hematopoietic cells which expressed GATA-1 , 2, and gamma, beta globin gene, and after being replated in methylcellulose medium, they formed typical erythroid colonies. Conclusion: Adult adipose tissue-derived Flk1^+CD31^-CD34^-- cells could differentiate into endothelial and hematopoietic cells. The results suggested that these Flk1^+CD31^-CD34^- cells bear characteristics of hemangioblast.
