摘  要:目的观察地塞米松对烧伤早期中性粒细胞体外凋亡的影响及bcl-2、核因子кB(NFкB)蛋白表达的变化。方法应用荧光染色和流式细胞仪观察地塞米松对正常血清和烧伤血清诱导烧伤早期中性粒细胞体外自发凋亡的影响;免疫组织化学SP法检测中性粒细胞bcl-2、NFкB蛋白表达。结果烧伤血清较正常血清诱导中性粒细胞凋亡减少(6.18±0.96vs.21.77±2.32,P<0.05),bcl-2、NFкB蛋白表达增加(83.27±5.45vs.49.95±2.67,0.1802±0.0130vs.0.0434±0.0196,P<0.05);加入地塞米松后,烧伤血清组凋亡增加(12.67±0.71vs.6.18±0.96,P<0.05);且NFкB蛋白表达减少(0.1031±0.0154vs.0.1802±0.0130,P<0.05),而正常血清组差异无显著性。结论地塞米松可减轻烧伤血清对中性粒细胞凋亡的抑制作用,其机制可能与NFкB蛋白表达下调有关。Objective To study the effect of dexamethasone on spontaneously apoptosis, bcl-2, arid neuclear facor KB (NFKB) expressions of polymorphonuclcar neutrophil (PMN) from postbum rabbits. Methods PMN were isolated from 8 rabbits on 24 posthum hours and cultured with normal serum (NS), bum serum (BS), normal serum plus dexamethasone (ND), and bum serum plus dexamethasone (BD) for 24 hours, respectively. The quantification of apoptosis was analyzed by acridine orange + ethidium bromide fluorescent staining and flow cytometry , and the contents of bcl-2 and NFKB protein detected by immunohistochemical method. Results In the KS group, the percentage of apoptotic PMN decreased (compared with NS group, 6. 18 ±0.96vs. 21.77 ±2.32, P〈 0. 05), and the contents of bcl-2 and NFKB protein increased (compared with NS group, 83. 27 ± 5.45 vs. 49. 95 ± 2. 67, P 〈0. 05 ). When compared with BS group, the apoptotic percentage of BD group increased (12. 67 ± 0. 71 vs. 6. 18 ± 0. 96, P 〈 0. 05 ), and the content of NFKB protein reduced (0. 1031 ± 0. 0154 vs. 0. 1802 ± 0. 0130, P 〈 0. 05), but no significant difference between ND and NS group was found. Conclusion Dexamethasone decreases the inhibition of PMN apoptosis by bum serum, which may be associated with the down-regulation of NFкB expression.
