氨基葡萄糖及硫酸软骨素对啮齿类动物的镇痛抗炎作用

摘　　要：[目的]观察氨基葡萄糖(glucosamine,GS)和硫酸软骨素(chondroitin sulfate,CS)以及两者联合使用时,对大鼠和小鼠的镇痛与抗炎作用。[方法]实验设3个试验组和1个空白对照组。3个试验组分别为A组(1g/kg体重GS),B组(0.5g/kg体重CS),C组(1g/kg体重GS+0.5g/kg体重CS)和D组(蒸馏水)。连续对小鼠和大鼠灌胃30d后,分别进行小鼠热板试验、扭体试验、耳廓肿胀试验和大鼠足跖肿胀试验。[结果]小鼠热板试验A组的平均痛反应发生时间最长,为20.28s、扭体试验C组的平均扭体反应次数最少,为12.64次、耳廓肿胀试验A组的平均耳廓肿胀度最小,为3.56mg;大鼠足跖肿胀试验4h时A组的平均足跖肿胀度最小,为0.36ml。提示GS、CS以及两者联合使用均能延长小鼠的痛反应发生时间;减少小鼠15min内的扭体次数、小鼠耳廓肿胀度以及大鼠4h时的足跖肿胀度。[结论]GS、CS以及两者联合使用对动物都具有镇痛和抗炎作用,并且三者的作用程度相似。[ Objective ] To observe the analgesic and anti-inflammatory effects of glucosamine （ GS ）, chondroitin sulfate （ CS）and the two in combination on mouse and rat. [ Methods ] Four groups were designed in the research. Mice and rats in group A were administrated by gavage with lg/kg＇bw GS, group B with 0.5 g/kg.bw CS, group C with 1 g/kg·bw GS ＋ 0.5 g/kg·bw CS, and in group D were treated with distilled water only as control. After 30-continuous-days＇ treatment, mouse hot plate, mouse twist, mouse ear edema and rat paw swelling tests were conducted to assess the analgesic and anti-inflammatory effects of GS and CS. [ Results ] In mouse hot plate test, group A had the longest average time of pain reaction which was 20.28 s. In mouse twist test, the least average twist frequency was 12.64 times found in group C. In mouse ear edema test, group A holded the lightest average degree of ear edema which was 3.56 mg, and also in group A the lightest average degree of swelling was 0.36 ml in rat paw swelling tests. GS, CS and a combination of both compounds can delay the time of pain reaction, decrease the frequency of mouse twist and reduce the degrees of ear edema in mice, and reduce the paw swelling in rats. [ Conclusion ] Both GS and CS, and their combination possess analgesic and anti-inflammatory effects.
