摘 要:[目的]探讨某中药口服液对汞染毒小鼠的治疗作用及剂量-效应关系。[方法]用60只小白鼠,数字表法随机分成4组(n=15):大剂量治疗组、小剂量治疗组、模型组和正常组。前三组用133mmol/L氯化汞溶液皮下注射染毒,连续30d。染毒14d后,用两个不同剂量0.2ml/只和0.05ml/只的某中药口服液分别给大、小剂量治疗组小白鼠灌胃,直至实验结束。模型组和正常组用等体积的蒸馏水灌胃。称实验前、后的体重,测定实验前、后的血红蛋白(Hb)含量。实验结束时从眼球后取血,分离血清,处死动物,取大脑,冰浴下匀浆。测定其血清尿素、丙氨酸氨基转氨酶(ALT)、胆固醇(TC)、大脑匀浆乙酰胆碱酯酶(TChE)活力、超氧阴离子自由基(O_2^(?))浓度。[结果]正常组小鼠实验后体重明显升高,与其他组比较,P<0.01;实验后小剂量治疗组和模型组的Hb含量明显低于实验前(P<0.01),同时也明显低于大剂量治疗组和正常组(P<0.01);血清ALT活性模型组明显高于其他三组(P<0.01);血清尿素正常组明显高于其他三组(P<0.01),小剂量治疗组明显低于其他三组(P<0.05和<0.01),模型组明显高于小剂量治疗组(P<0.05);胆固醇,模型组明显高于小剂量治疗组(P<0.05);大脑TChE活力,各组差异无显著性意义(P>0.05);各实验组脑匀浆对O_2^(?)清除率依次是:正常组>大剂量治疗组>小剂量治疗组>模型组(P<0.01)。[结论]大剂量某中药口服液对汞染毒小鼠具有升高血液Hb、脑对O_2^(?)清除率作用以及降低血ALT、脑O_2^(?)浓度的作用。[ Objective ] To investigate the curative effects of Haierfu oral liquid on mercury poisoned mice and its dose -effect relationship. [ Methods ] Sixty mice were divided into four groups, high-dosage treated group, low-dosage treated group, model group, and normal control group. The former three groups were given with 133mmol/L of mercuric chloride ( HgC12 )by hypodermal injection daily for 30 days. After two weeks of mercury poisoning, mice in high-dosage and low-dosage treated groups were given with 0.2 ml and 0.05 ml of Haierfu oral liquid respectively by gastric perfusion. Mice in model and normal control groups were perfused with distilled water only. Body weight and Hb of all animals were measured before and after the experiment. At the end of the experiment, urea, ALT and TC content in serum as well as TChE activity and the level of O2 in the brain were determined. [ Results ] In contrast to others, the weight of normal control group increased significantly( P 〈 0.01 ). Serum Hb in model group and low-dosage treated group were significantly lower than that in normal control group and high-dosage treated group ( P 〈 0.01 ).Serum ALT activity in model group was significantly higher than that in other three groups ( P 〈 0.01 ). Serum urea, in normal control group was significantly higher than that in other three groups ( P 〈 0.01 ), in low-dosage treated group was lower than that in other three groups( P 〈 0.05 and 〈 0.01 ), and in model group was significantly higher than that in low-dosage treated group ( P 〈 0.05 ). TC in model group was significantly higher than that in low-dosage treated group( P 〈 0.05 ). No significant difference of TChE activity in brain was observed among all groups ( P 〉 0.05 ). The clearance rate of O~ in brain in different groups was decreasing in the order of normal control group 〉 high-dosagee treated group 〉low-dosage treated group 〉model group( P 〈 0.01 ).
