摘 要:目的探讨脂质载体前列腺素E1(Lipo-PGE1)对皮肤缺血-再灌注损伤压疮大鼠模型的干预作用,为压疮的防治提供依据。方法将24只大鼠随机分成缺血组(I组)、缺血-再灌注组(IR组)、Lipo-PGE1干预组(PGE1组)各8只。三组均于压疮装置上受压2h,I组解除受压后立即处死;IR组、PGE1组解除受压前10min分别于尾静脉注射生理盐水、Lipo-PGE1,解除受压4h时处死。结果与I组、PGE1组比较,IR组SOD活性显著降低,MDA、LDH、ET-1、NO含量显著增加(均P<0.05)。而PGE1组各项指标与I组比较,差异无显著性意义(均P>0.05)。结论缺血-再灌注损伤在压疮形成中发挥作用,Lipo-PGE1能有效拮抗氧自由基对皮肤的损伤,减轻皮肤缺血-再灌注损伤。Objective To evaluate the effects of Lipo-PGE1 on the pressure ulcers involved ischemia-reperfusion injury in rats. Methods twenty-four rats were classified into 3 groups randomly: the ischemic group (group Ⅰ) ; the ischemiareperfusion group (group IR) ;Lipo-PGE1 treated group (group PGE1 ). All rats were subjected to decubitus compression model for 2 hours. Group Ⅰ were sacrificed right after decompression, while group IR and group PGE1 were treated with injection of normal saline and Lipo-PGE1 respectively 10 minutes before decompression and sacrificed 4 hours after decompression. The activity levels of superoxide dismutase (SOD), lactic dehydrogenase (LDH) and the contents of malondialdehyde (MDA), endothelin-1 (ET-1) and nitric oxide (NO) were all measured. Results The activity level of SOD in group IR was lower than that in group I and group PGE1 ; while the contents of MDA, LDH, ET-1 and NO were significantty increased(all P〈0.05). There were no significant differeces of the indexes between group PGE1 and group Ⅰ (all P〉0.05). Conclusion Ischemia-reperfusion injury played a vital role in the development of pressure ulcer. Lipo-PGE1 can be effectively against the skin injury by oxygen free radical, and relieve the ischemia-reperfusion injury of skin.
