摘  要:目的探讨龙胆抗病毒有效部位(RG3-1)体外抑制呼吸道合胞病毒(RSV)的作用。方法采用煎煮法、二氯甲烷萃取法和柱层析法分离提取RG3-1。以病毒唑为阳性对照药物,采用细胞病变抑制试验观察RG3-1对RSV的抑制作用。结果RG3-1半数中毒浓度(TC50)为5.29mg/mL,抑制RSV的半数有效浓度EC50为262.95μg/mL,治疗指数(TI)为20.12;RG3-1对RSV的抑制存在量效关系;RG3-1具有直接灭活RSV的作用;在感染后至少10h以内给药,RG3-1能够有效的抑制RSV复制。结论RG3-1在HeLa细胞中对RSV有明显的抑制作用。Objective To study the anti-respiratory syncytial virus (RSV) effect of an compound (RG3-1) from radix gentianae in vitro .Methods RG3-1 was extracted from radix gentianae by hot water, dichloromethane, and column chromatography. The cytotoxicity and antiviral action of RG3-1 were tested by cytopathic effect (CPE) inhibition assay,and ribavirin as the positive control drug. Results It showed good inhibitory effect against RSV with the medium cytotoxicity concentration (TCso) 5.29mg/mL, the medium effective concentration (ECso) 262.95μg/mL,and the treatment index(TI) 20.12. RG3-1 was effective antiviral agent for RSV in a concentration-dependent manner. RG3-1 also displayed direct inactivation on RSV. It also exhibited obvious inhibition on RSV replications when RG3-1 was added in 10h after infection (P 〈 0.01 ). Conclusion RG3-1 can significantly inhibit RSV replications in HeLa cells.
