摘  要:目的为了解新疆地区结核分支杆菌耐异烟肼(INH)分离株Kat G基因突变情况,探讨其在INH耐药性中的可能作用。方法采用PCR和PCR-SSCP方法对60株结核分支杆菌临床分离株Kat G基因突变进行检测。结果以结核分支杆菌H37RV为对照,28株药物敏感株的KatG基因PCR扩增产物273bp片段28例SSCP图谱正常。32例耐INH分离株中KatG基因扩增均阳性,其中11株SSCP图谱异常,其敏感性为34.37%。结论新疆地区结核分支杆菌INH耐药株存在KatG基因突变,其突变对INH耐药性的影响有待于进一步研究。Objective To identify if there is correlation between INH resistance and KatG mutation of Mycobaterium tuberculosis in Xinjiang region and the mechanism of INH tolerance of Mycobaterium tuberculosis in Xinjiang at molecular level. Methods There 60 clinical isolates of Mycobacterium tubeculosis (32 isolates was resistant to drugs and 28 sensitive to drugs ) were detected by using PCR and PCR - SSCP. Results SSCP pattern of Mycobacterium tubeculosis H37RV was as contral, SSCP patents of KatG gene with the size od 273bp amplified by PCR from 28 sensitive isolates were same as that of control. There the SSCP pattern of 11 isolates out of 32 isolates expressing KatG gene and reislstant to drugs were abnormal with a mutation rate of 34.37 %. Conclusion The occurrence of drug resistance of INH isoaltes of Mycobacteriu tuberculosis in Xinjiang might be due to mutation of KatG gene and mechanisms of drug resistance should be further investigated on molecular level.
