托吡酯、拉莫三嗪对大鼠学习与记忆功能影响的比较研究

摘  要:目的研究抗癫痫药物对大鼠认知功能的影响。方法健康雄性性成熟SD大鼠40只,随机分为正常对照组(NS)、癫痫对照组(PTZ)、托吡酯(TPM)组及拉莫三嗪(LTG)组,每组10只。除NS组外,其他3组用PTZ点燃。治疗2周后用莫礼斯(Morris)水迷宫测试认知功能。采用方差分析进行统计学检验。结果PTZ组学习成绩提高很快(P﹤0.05,P﹤0.01)。LTG组测试成绩提高很快(P﹤0.01),成绩优于NS组。TPM组测试成绩提高慢,无显著性差异,较NS组差。在寻找平台象限和在平台象限逗留时间的测试中,TPM组寻找时间最长,逗留时间最短,两项成绩均不如其他组(P﹤0.05,P﹤0.01),其他各组无显著性差异。结论TPM可影响大鼠的学习、记忆、判断功能,LTG能提高大鼠的学习、记忆、判断功能。To explore the effectiveness of topiramate and lamotrigine on the cognition of rats. Methods Forty adolecent male SD rats, weighted (210+23)g, were divided into 4 groups randomly:normal control group (NS group), status epileptics group (PTZ group), topiramate(TPM) treatment group and lamotrigine(LTG) treatment group. Except the NS group, all rats were kindled by PTZ, followed by treated with topiramate and lamotrigine in two groups. Two weeks later, 4 groups were tested with Morris water maze. Results The performance record of the PTZ group, the LTG group had been improved quickly (P 〈 0.05, P 〈 0.01), and was better than that of NS group. The performance record of TPM group was increased slowly (P 〉 0.05), and was worse than that of the NS group. The time of TPM group was longer than other groups in searching for platform quadrant (P 〈 0.01)but was shorter than that of the LTG group(P 〈 0.01) in stay at the platform quadrant. No difference was found between in other groups. Conclusions TPM impairs the learning ability of rats' some function of cognition and memory, but LTG can improve them.
