姜黄素治疗大鼠非酒精性脂肪肝病的实验研究

摘　　要：目的探讨姜黄素治疗大鼠非酒精性脂肪肝病(NAFLD)的治疗作用。方法42只SD雄性大鼠分为正常对照组(14只)和NAFLD组(28只),以高脂饲料饲养建立NAFLD大鼠模型,将其随机平分为两组:NAFLD对照组和姜黄素治疗组,12周末,处死所有动物,检测血脂、肝功能、肿瘤坏死因子α(TNF-α)、脂联素、肝脂质等指标,并观察肝组织病理学变化。结果姜黄素治疗组与NAFLD模型对照组比较,肝功能明显改善,谷草转氨酶AST(u/L)146±27与254±81,谷丙转氨酶ALT(u/L)48±24与106±41(P<0.01);胰岛素抵抗指数明显降低(HOMA-IR),分别为6.5±1.8与10.3±3.1(P<0.05);TNF-α明显降低,分别是132.7±26.9与316.2±36.1(P<0.01);脂联素水平明显升高,分别是20.6±2.5与14.3±0.9(P<0.05);肝TG、TC、LDL-C水平显著减低,分别是1.51±0.35与2.17±0.63、0.655±0.68与1.235±0.79、0.29±0.30与0.73±0.52(P<0.05);肝脂肪变性明显好转。结论姜黄素可以用来治疗大鼠NAFLD,其机制可能是通过抑制JNK酶的活性来起作用。Objective To study the effects of curcumin treatment on rat non-alcoholic fatty liver disease（NALFD）. Methods Forty-two male SD rats were randomized into 2 groups,normal control group（14 cases） and NAFLD group（28 cases）. To establish the NAFLD rat model, And NAFLD model were subdivided into 2 subgroups： NAFLD control group and curcumin treatment group. At the end of 12 week, all rats were killed to detect the blood lipids, liver function, TNF-α,adiponectin, lipids of the liver, etc. And the pathological changes of liver organ were observed. Results Liver function was significantly improved in curcumin treatment group compared with NAFLD control group. AST was 146±27 u/L vs 254＋81 u/L;ALT was 48±24 u/L vs 106±41, respectively（P〈0.01） ; HOMA-IR and TNF-α were significantly lower,but adiponectin was hgher, in curcumin treatment group than that of NAFLD control group. HOMA-IR was 6.5±1.8 vs 10.3±3.1 （P〈0.05） ;TNF-α was 132.7±26.9 vs 316.2±36.1 （P〈0. 01）;Adiponectin was 20. 6±2.5 vs 14.3±0. 9 （P〈0.05）. The liver TG,TC,and LDL-C were all significantly lower in curcumin treatment group than that of NAFLD control group. The liver TG,TC,and LDL-C were respectively 1.51 ±0.35 vs 2.17 ±0.63,0. 655±0.68 vs 1. 235±0.79,0.29±0.30 vs 0.73±0.52 （P〈0.05）. The fatty degeneration of liver was significantly improved. Conclusion Curcumin can be used to treat NAFLD through the inhibition of JNK acitivity.
