小大黄对ox-LDL诱导血管平滑肌细胞增殖的影响

摘　　要：目的:观察小大黄对氧化修饰型低密度脂蛋白(ox-LDL)诱导血管平滑肌细胞(SMC)增殖的影响。方法:采用体外培养的牛主动脉平滑肌细胞为模型,ox-LDL诱导SMC增殖。实验分为对照组、ox-LDL组和用药组(ox-LDL+不同剂量小大黄),应用MTT法、流式细胞仪分别测定细胞数量和细胞周期,用硫代巴比妥酸比色法测定细胞培养液中丙二醛(MDA)的水平。结果:小大黄能明显降低细胞培养液中MDA的水平,MTT检测显示用药组比ox-LDL组OD值降低,并呈浓度依赖性。流式细胞仪检测表明,用药组(S+G2M)期细胞百分率降低。结论:小大黄能抑制ox-LDL诱导的SMC增殖,并具有抗脂质过氧化作用。Objective:To observe the effect of polygonum sibiricum laxm on vascular smooth cells proliferation induced by oxidized LDL.Methods:The subjects were divided into 5 groups:control group,ox-LDL group and three polygonum sibiricum laxm treatment groups.The experimental models of proliferation of cultured bovine VSMC induced by oxidized-low density lipoprotein,the MDA level was determined with thiobarbituric acid reactive substance and the number of bovine VSMC was tested with MTT and the cell cycle phase was determined by flow cytometer.Results:Polygonum sibiricum laxm could obviously reduce the level of MDA in the medium and inhibit the proliferation of VSMC in a concentration-dependent fashion.Polygonum sibiricum laxm could reduce the(S+G2M) block in cell cycle traverse.Conclusion:Polygonum sibiricum laxm can inhibit the proliferation of VSMC,which maybe related to the inhibition of lipid peroxidation.
