摘　　要：目的探讨粒系集落刺激因子(G-CSF)作为造血干细胞动员剂对健康供者外周血单核细胞及NK细胞表型的影响。方法对25名自愿捐献的健康供者应用G-CSF作造血干细胞动员,分别于动员前、动员后(干细胞采集当日)、干细胞采集后7d及采集后1个月收集健康供者的外周血标本,用流式细胞仪检测细胞数量及细胞免疫表型。结果动员后外周血单核细胞和NK细胞的绝对数量分别动员前增高4.96和2.02倍,采集后7d二者都恢复到动员前水平。动员前后单核细胞表面CD14的抗原表达的差异无统计学意义(P>0.05),但HLA-DR抗原表达强度明显减弱(P<0.05),于采集后7d恢复到动员前水平;动员前后NK细胞表面CD16抗原表达差异无统计学意义(P>0.05)。结论G-CSF作为造血干细胞动员剂可以使健康供者外周血的单核细胞和NK细胞数量在短期内增加;使单核细胞表面HLA-DR抗原表达在短期内减弱,但不影响单核细胞表面CD14抗原及NK细胞表面CD16抗原的表达。Objective To study the effect of administration of granulocyte colony-stimulating factor （G-CSF） on the phenotype of monocytes and natural killer （NK）cells in healthy individuals during mobilization of hematopoietic stem cells. Methods Peripheral blood samples from 25 healthy donors were taken prior to, after mobilization （the day of stem cell collection） by G-CSF, on day 7 and 1 month after the collection of stem cells. Flow cytometry was used to assay the amount and intensity of antigen expression on monocytes and NK cells. Results After mobilization, the absolute count of monocytes and NK cells increased by 4.96 and 2.02 folds, respectively, but returned to pre-mobilization levels on the day 7 after stem cell collection. The antigen expression of HLA-DR on monocytes decreased （ P〈0.05） and returned to pre-mobilization levels 7 days later. The CD14 expression on monocytes and CDI 6 on NK cells showed no significant change after mobilization. Conclusion G-CSF could induce increase of monocytes and NK cells as well as decrease of HLA-DR expression of monocytes in a short term after mobilization. No significant change of antigen expression of CD14 on monocytes and CDI 6 on NK cells were found.
