摘  要:目的建立反相高效液相色谱法同时测定人血浆中10种苯二氮类药物(氯氮、咪达唑仑、硝西泮、奥沙西泮、艾司唑仑、劳拉西泮、阿普唑仑、氯硝西泮、地西泮和三唑仑)。方法血浆样品经二氯甲烷萃取后,采用Shim-pack CLC-C8色谱柱,柱温35℃,流动相为0.01mol.L-1磷酸盐缓冲液-乙腈-无水乙醇-甲醇(68.2∶31.2∶0.3∶0.3),流速为1.10mL.min-1,紫外检测波长230nm。结果血浆中氯氮、咪达唑仑、硝西泮、奥沙西泮、艾司唑仑、劳拉西泮、阿普唑仑、氯硝西泮、三唑仑和地西泮在40~2000μg.L-1内线性关系良好(r≥0.9992),平均方法回收率均在97%~104%,萃取回收率均在70%以上,日内和日间RSD均≤11%(n=5)。结论本方法准确快速、简便易行,适用于人血浆中10种苯二氮类药物的同时检测。OBJECTIVE To establish a HPI,C method for simultaneous determination of frequently prescribed 10 benzodiazepines ( chlordiazepoxide, midazolam, nitrazepam, oxazepa, estazolam, lorazepam, alprazolam, clonazepam, diazepam and triazolam) in human plasma. METHODS The benzodiazepines were extracted with dichloromethane from plasma samples, and then was analyzed on a Shim-pack CLC-Cs column(6. 0 mm× 150 mm,5 μm) at 35 ℃ with the mobile phase consisting of 0. 01 mol · L^-l phosphate buffer solution-acetonitrile-ethanol-methanol (68.2:31.2: 0. 3:0. 3), at a flow rate of 1.10 mL · min^-1. Ultraviolet detection wavelength was at 230 nm. RESULTS The linear ranges were 40 -2 000 μg · L^-l (r≥0. 999 2). The average recovery of 10 drugs were within 9 7% - 104%. The average extraction recoveries were all over 70%. Intra-day RSD and inter-day RSD were less than 11% (n = 5 ). CONCLUSION The method is accurate,rapid and convenient for simultaneous determination of 10 benzodiazepines in human plasma.
