摘  要:目的做为中国急性弛缓性麻痹(AFP)病例监测系统和中国维持无脊髓灰质炎(脊灰)状态的一部分,对日喀则地区2005年健康儿童肠道病毒(EV)的携带情况进行监测分析,为西藏自治区维持无脊灰提供实验室依据。方法2005年3月在日喀则地区3个县的0~5岁健康儿童中共采集粪便标本70份,进行EV分离和血清型鉴定。对分离到的脊灰病毒(PV)同时使用逆转录-聚合酶链反应和酶联免疫吸附试验进行型内鉴定,对分离到的所有非脊灰肠道病毒(NPEV)进行VP1区核苷酸序列测定以鉴定血清型。结果从70份粪便标本中共分离到16株PV,阳性率为22.86%。型内鉴定及VP1区核苷酸序列测定和分析结果显示,2005年日喀则地区无脊灰野病毒和疫苗衍生脊灰病毒的存在;共分离到9株NPEV,阳性率为12.86%。结论日喀则地区2005年继续保持无脊灰状态。EV监测为后续研究提供宝贵的资料,也为EV监测逐渐整合到AFP病例监测系统中做了有益的尝试。Objective As a part of Chinese AFP surveillance system and the activity of maintaining polio-free in China,the carrying situation of enterovirus in healthy children in Rikaze of Tibet was analyzed,which applied the laboratory evidence for maintaining polio-free in Tibet. Methods A total of 70 stool samples were collected from healthy children aged less than 5 years in 4 countries of Tibet RiKaze in March,2005. Enterovirus was isolated from these stool samples and then was serotyped by neutralization using specific antisera. All the polioviruses isolated were screened by RT-PCR-RFLP and ELISA,and VP1 code gene of all the non-poliovirus enterovirus (NPEV) isolated were sequenced in order to identify serotyping. Results Sixteen polioviruses were isolated from 70 stool samples and carrying rate of poliovirus was 22.86%. The results of intratypic differentiation (ITD) method and VP1 region sequencing show that there was not wild poliovirus and vaccine derived poliovirus (VDPV) in Tibet in 2005. Nine NPEV were isolated from the 70 stool samples and the carrying rate of NPEV was 12. 86%. Conclusion This report analyzed the carrying situation of enterovirus in healthy children in Rikaze of Tibet and confirmed polio-free status in the area. NPEV surveillance will provide the consequential study of enterovirus with valuable information and be a profitable try for integrating enterovirus surveillance into AFP surveillance system.
