摘 要:【目的】观察补肾健脾中药助孕3号方对肾虚黄体不健流产大鼠T淋巴细胞抗原识别受体(TCRγδT)的影响。【方法】将SD大鼠40只随机分为5组,即空白对照组,模型组,助孕3号方高、低剂量组(中药高、低剂量组,剂量分别为7.08、3.54 g.kg-1.d-1),多力玛组(剂量为1.5 mg.kg-1.d-1);除空白对照组外,其他组均采用羟基脲(剂量为450 mg.kg-1)联合米非司酮(剂量为3.75 mg.kg-1)复制肾虚黄体不健流产模型。观察各组大鼠的子宫质量、胚胎数及流产情况,采用流式细胞仪检测外周血TCRγδT及CD3+T淋巴细胞水平。【结果】模型组大鼠子宫质量、孕胎数显著性降低,流产率显著性升高(P<0.01);中药高、低剂量组均可显著性升高子宫质量及孕胎数,降低流产率(P<0.05或P<0.01),且呈现一定的量效关系,其作用与孕激素多力玛相似。模型组CD3+T细胞水平显著性升高,TCRγδT细胞/CD3+T细胞比值显著性降低,TCRγδT细胞水平有降低趋势,中药高剂量组可显著性升高TCRγδT细胞水平和TCRγδT细胞/CD3+T细胞比值,低剂量组可降低CD3+T细胞水平(均P<0.05或P<0.01)。【结论】助孕3号方的保胎作用可能与其纠正免疫失衡状态及降低流产率有关。[ Objective] To observe the effect of Zhuyun Prescription Ⅲ (ZPⅢ), which is composed of kidneytonifying and spleen-strengthening herbal medicine, on T lymphocyte antigen recognition receptors (TCRγδT) in luteal insufficiency abortion rats with deficiency of kidney. [ Methods ] Forty SD rats were equally randomized into 5 groups: blank control group, model group, high- and low-dose ZPⅢ groups (in the doses of 7.08 and 3.54 g·kg^-1 . d^-1, respectively) and allylestrenol tablets ( 1.5 mg·kg^-1·d^-1 ) group. Luteal insufficiency abortion rats model with deficiency of kidney was established by hydroxyurea (450 mg·kg^-1 ) and mifepristone (3.75 mg·kg^-1). The uterus weight, the amount of embryo and the abortion rate were observed, and the peripheral blood levels of TCRγδT and CD3^+T lymphocytes were detected with flow cytometer. [ Results ] The uterus weight and the amount of embryo were decreased , and the abortion rate was increased in the model group (P 〈 0. 01 ) ; high- and low-dose ZPⅢ counteracted the above changes in the model group (P 〈0. 05 or P 〈0. 01 ) in dose-dependent manner, and the effect was similar to that of allylestrenol tablets. The CD3^+T lymphocytes increased , the ratio of TCRγδT/CD3^+T lymphocytes decreased and TCRγδT cells level reduced in the model group. High-dose ZPⅢ increased the TCRγδT cells level and the ratio of TCRγδT/CD3^+T lymphocytes, while low-dose ZPⅢ decreased CD3^+T lymphocytes level (P 〈0.05 or P〈0.01). [Conclusion] The anti-abortion mechanism of ZPⅢ is probably related to the adjustment of immune imbalance and the decrease of abortion rate.
