摘 要:目的比较从乳猪肝中提取促肝细胞生长素溶液的不同方法。方法对乳猪肝匀浆液分别用反复冻融、酸水解和酶水解3种方法提取,计算单位收率及单位生产成本。结果3种方法制备的促肝细胞生长素溶液均符合国家药品标准,但收率和活性不同。用酸水解法,多肽含量及收率比反复冻融法提高2倍,酶解法多肽含量及收率比反复冻融法提高2.4倍,酸解法和酶解法的活性值比反复冻融法都有所降低。酸水解法的单位成本只有反复冻融法的1/2,酶解法的单位成本与反复冻融法的基本相同。结论3种方法制备的促肝细胞生长素溶液在收率和成本方面有较大的不同,酸水解方法更适合于大规模生产。Objective To compare the methods to prepare the solution of promoting hepatocytes growth factor(pHGF), Methods The solution of pHGF was extracted from the suckling pig liver homogeuate by using the three methods of repeat freeze thawing, acidolysis and enzymic hydrolysis. The yield and cost of their unit production were evaluated. Results The pHGF solutions of prepared from the three methods were all consistent with the nation drug standard, but their yield and activity were different. While compared with that of the repeat freeze thawing, the yield and content of the solution increased twice by acidolysis and 2.4 times by enzymic hydrolysis increased. But their biological activity values slightly decreased as compared with that of the repeat freeze thawing. The unit production cost of the solution prepared by the acidolysis was only half as that of the repeat freeze thawing, and that of the enzymic hydrolysis was the same as that of the repeat freeze thawing. Conclusion The three methods of the preparation of the pHGF solution had a big different in the yield and the production cost. The acidolysis is suitable to the large scale production of pHGF solution.
