摘  要:目的:研究支气管哮喘(以下简称哮喘)大鼠神经内分泌免疫网络(NEI)若干指标的变化;观察补肾药淫羊藿对哮喘大鼠NEI网络若干指标的影响。方法:健康雄性挪威(BN)大鼠54只,分为正常对照组(A组),哮喘对照组(B组),低剂量淫羊藿组(C组),中剂量淫羊藿组(D组),高剂量淫羊藿组(E组)。用实时定量-PCR(Realtime-PCR)法检测下丘脑促肾上腺激素释放激素(CRH)mRNA的表达;放免法检测血浆中促肾上腺皮质激素(ACTH)的含量;酶联免疫吸附实验(ELISA)检测血清IL-4,IL-6,IFN-γ和皮质酮(CORT)的含量。结果:哮喘大鼠下丘脑CRHmRNA表达和血浆ACTH无明显变化,血清皮质酮水平升高显著,细胞因子IFN-γ无明显变化,IL-6、IL-4有上升趋势,低、中剂量淫羊藿能够促进哮喘大鼠CRHmRNA的产生,中剂量淫羊藿能够上调血清IFN-γ的浓度。结论:哮喘大鼠存在NEI网络的紊乱;补肾药淫羊藿可以增强下丘脑-垂体-肾上腺皮质(HPA)轴的功能,提高血清IFN-γ的水平。这些可能是其治疗哮喘的机制之一。Objective:To study changes of several related subjects of neuro-endocrino-immune network(NEI) in asthma models, to study the effects of Epimedium Herb on NEI. Methods: 54 BN rats were randomly divided into 5 groups, Group A, B, C, D and E. Group A was the normal group. Group B was the allergic asthma model. Group C, D and E were administered respectively with low, moderate and high dose Epimedium Herb. CRHmRNA, ACTH, CORT, IL-6, IL- 4, IFN-γ were measured in all groups. Results:The expression of CRHmRNA in hypothalamic tissue and concentration of interferon gamma in blood both increased in controlled groups in comparison with group B. There were higher expression of CRHmRNA in group C or D than that in group B. There were higher concentration of IFN-γ in group D than that in group B. Conclusion:The rats troubled with repeated asthmatic attack for three weeks had disorders of NEI( mainly including HPA axis and several cytokines). Epimedium Herb could improve HPA axis and Th1/Th2 cytokines so as to alleviate the inflammation of asthma.
