摘  要:目的探讨p-ERK1/2在血管紧张素-(1-7)(Ang-(1-7))抑制血管紧张素Ⅱ(AngⅡ)诱导的大鼠肾系膜细胞(GMC)增殖中的作用。方法在培养的GMC中,加入不同浓度的Ang-(1-7)与Ang-Ⅱ共同培养,采用结晶紫计数法检测GMC数目变化;western blot检测GMC中p-ERK1/2蛋白表达变化。结果Ang-(1-7)呈剂量依赖性地抑制Ang-II诱导的GMC数目的增加和GMC内p-ERK1/2蛋白的表达。结论ERK通路参与了Ang-(1-7)抑制Ang-II诱导的GMC的增殖。[Objective] To investigate the effect of p-ERK1/2 during angiotensin-(1-7) inhibiting proliferation of rat glomerular mesangial cells (GMC) induced by angiotensin Ⅱ. [Methods] GMCs were co-cultivated with angiotensin Ⅱ and different dose of angiotensin-(1-7). GMC numbers were evaluated by crystal violet staining and the amount of p-ERK1/2 expression were detected by western blot. [Results] Angiotensin-(1-7) showed its inhibitory effects on GMC number increasing induced by angiotensin Ⅱ as well as the amount of p-ERK1/2 expression in a concentration- dependent manner. [Conclusion] ERK singal pathway is involved in the inhibiting effects of angiotensin-(1-7) on angiotensin Ⅱ -induced GMC proliferation.
