摘  要:目的研究黄芪对高糖所诱导的血管内皮细胞损伤的保护作用。方法采用人脐静脉血管内皮细胞株EVC-304,在培养液中加入不同浓度的黄芪,孵育24h,高糖诱导损伤,再培养4h后,终止培养。观察细胞形态学变化并检测细胞活力、蛋白质含量,检测VEGFmRNA及蛋白的表达。结果正常对照组及黄芪各剂量组细胞活力、蛋白质含量均高于高糖损伤对照组,差异有显著性(P<0.05)。高糖明显抑制VEGF的活性和表达,黄芪则有明显的修复作用。结论黄芪可以减轻高糖对血管内皮细胞的损伤,具有一定保护作用。[Objectiove] To study protective effect of astragalus on vascular endothelial ceils which were damaged by glucose. [Methods] The damaged insult model of human umbilical vein endothelial ceils (VEC-304) were induced by glucose for 4 h. The cells were pretreated with three concentrations of astragalus solution for 24 h, before they were exposed to glucose. MTT and Protein levels in ceils were determined after 4 h. [Results] MTT and protein levels in ceils in normal control group and astragalus treated groups were higher than in glucose damaged group without glucose (P 〈0.05.) The expression of VEGF was inhibited by glucose. [Conclusion] Astragalus can protect th injury of vascular endothelial ceils following glucose.
