摘  要:目的探讨丹参注射液对单侧输尿管梗阻(UUO)大鼠肾间质纤维化的影响。方法采用UUO致肾间质纤维化大鼠模型。24只雄性Wistar大鼠,随机分为假手术组、模型组和丹参组。术后第10天取术侧肾组织做HE,Masson染色及转化生长因子(TGF)β-1,α-平滑肌肌动蛋白(α-SMA)免疫组织化学检测。结果丹参组显著改善梗阻侧肾组织病理变化,下调TGFβ-1和α-SMA蛋白表达。结论丹参注射液对梗阻性大鼠肾间质纤维化有保护作用。Objective To study the mechanism of the effects of Salvia Miltiorrhiza injection on renal interstitial fibrosis caused by unilateral ureteral obstruction (UUO) in rats. Methods UUO rat models were used. Twenty-four male wistar rats were randomly divided into three groups with 8 rats each;sham-operated group (A), UUO group (B) and Salvia Miltiorrhiza injection treated group (C). After 10 days of UUO, rats were killed and the operated kidneys were collceted for HE, Masson staining and immunohistochemical staining fo TGF-β1, α-SMA. Results In obstructed kidneys, there were significant interstitial fibrosis and interstitial space expansion. The relative volume of cortical interstitium significantly increased (P 〈 0.01 ). Immunohistochemical staining showed that TGF-β1, α-SMA expression remarkably increased in the obstructed kidneys (P〈0.01). In comparison with the model group, Salvia Mihiorrhiza Injection significantly decreased interstitial volume and the expression of TGF-β1, α-SMA ( P 〈 0.01 或 P 〈 0.05). Conclusion Salvia Miltiorrhiza Injection can down-regulate the expression of TGF-β1, α-SMA in obstructed kidneys. It can ameliorate pathological lesion and prevent progre.ss of renal interstitial fibrosis caused by UUO in rats.
