摘 要:目的比较磁导向下PEG修饰的5-氟尿嘧啶磁性白蛋白微球(PEG-5-Fu-MAMS)和5-氟尿嘧啶磁性白蛋白微球(5-Fu-MAMS)对胰腺的磁靶向性,以寻求一种对胰腺癌的磁导向化疗更有效的新途径。方法取Wistar大鼠18只,每组6只,分为5-Fu组、5-Fu-MAMS组和PEG-5-Fu-MAMS组;分别将三种不同的制剂(按5-Fu 8mg/kg)经胃左动脉注入胃十二指肠动脉,进入循环。并在胰腺表面加一表面强度为4250GS的磁场,30min后,处死大鼠取胰腺。用HPLC法检测其药物浓度。结果PEG-5-Fu-MAMS组胰腺中的药物浓度为76.18±4.09μg/ml;5-Fu-MAMS组为57.95±6.39μg/ml;5-Fu组为16.58±3.19μg/ml。PEG-5-Fu-MAMS组的药物浓度明显高于5-Fu-MAMS组和5-Fu组,差异具有显著统计学意义(P<0.01);5-Fu-MAMS组的药物浓度明显高于5-Fu组,差异具有显著统计学意义(P<0.01)。结论PEG修饰的磁性白蛋白微球具有更好的磁靶向性和亲水性,为实现胰腺癌的主动靶向治疗提供了一种新途径。Objective To compare the targeting distribution of PEG - 5 - Fu- MAMS and 5 - Fu - MAMS in pancreas under magnetic field and explore a better magnetic targeting chemotherapy for pancreatic cancer. Methods Eighteen mouse were equally divided into Group 5 - Fu (n = 6), Group 5 - Fu- MAMS (n = 6) and Group PEG- 5 -Fu- MAMS (n = 6). They were injected through the gastroduodenal artery at the dose of 8mg/kg of free drug, respectively.The pancreases were exposed in the magnetic field through an abdominal midline incision for 30 min. The animals were immediately killed after removing the magnetic field. The concentration of 5- fluorouracil in pancreas was determined by the high performance liquid chromatography (HPLC) . Results The 5 - Fu concentration in pancreas was 16.58 ± 3.19μg/ml, 57.95 ± 6.39μg/ml and 76.18 ±4.09μg/ml in Group 5 - Fu, 5 - Fu - MS and PEG - 5 - Fu - MS, respectively. There were significant difference between Group 5 - Fu and Group 5 - Fu - MS, Group 5 - Fu and Group PEG - 5 - Fu - MS, Group 5 - Fu - MS and Group PEG - 5 - Fu- MS (all P values, 〈 0.01). Conclusion PEG - 5 - Fu - MAMS could be efficiently delivery 5 - Fu to the targeted site compared with 5 - Fu- MS and free drug.
