摘  要:目的:评估瑞福纳米人工骨(胶原基纳米骨,nano-hydroxyapatie/collagen,nHAC)作为软骨组织工程细胞支架材料的可行性。方法:选择骨性关节炎(OA)患者行髋关节置换手术后外观大致正常的关节软骨,消化、分离、体外培养扩增后种植到nHAC中,复合物体外培养后进行Ⅱ型胶原免疫组织化学检测和扫描电镜观察。结果:软骨细胞可以长入nHAC支架材料中。同期软骨细胞保持了正常的外形且可分泌Ⅱ型胶原。结论:nHAC可以作为软骨细胞的载体。Objective To evaluate the value of as a scaffold for cartilage tissue engineering. Methods hip arthroplasty of osteoarthritis was selected, and then amplificated in vitro. The chondrocytes were seeded onto the nano-hydroxyapatite/collagen ( nHAC ) The normal cartilage from patients after total chondrocytes were digested, separated, and the nHAC scaffold and were cultured in a 3- dimensional environment for 5 and 10 days. The effects of the composite scaffold on the cell adhesion, proliferation, morphological changes, and synthesis of the extracellular matrix were observed by scanning electronic microscopy and immunohistochemistry. Results The chondrocytes could adhere to the surface of the scaffolds, proliferate, and migrate into the scaffolds. They maintained round and could secrete extracellular matrix on the porous scaffold. Conclusion The nHAC can be used as the cartilage cell carrier.
