摘  要:目的:探讨血管生成素受体Tie-2在糖尿病大鼠肾组织中的表达以及黄芪对其影响。方法:SD大鼠随机分为正常对照组、糖尿病组和黄芪治疗组。8周后处死大鼠,采用实时定量聚合酶链反应和免疫组化法分别检测3组大鼠肾组织中Tie-2基因和蛋白表达水平。结果:与正常对照组相比,糖尿病组大鼠肾组织Tie-2蛋白表达显著升高(P<0.01);黄芪治疗组大鼠肾组织Tie-2蛋白表达较糖尿病组显著下调(P<0.01)。结论:黄芪通过下调糖尿病大鼠肾组织Tie-2的表达而发挥肾保护作用是其治疗糖尿病肾病的作用机制之一。Objective: To study the expression of angiopoietin receptor Tie-2 in the renal tissue of diabetic rats and the effects of Astragalus. Methods: SD rats were randomly divided into normal control group, diabetes group and Astragalus-treated group. The expression of receptor Tie-2 in the renal tissue was assessed by using real-time quantitative polymerase chain reaction and immunohistochemical method. Results: Glomerule Tie-2 protein expression was significantly elevated in the diabetes group as compared with the normal control group (P〈0.01). Glomerule Tie-2 protein expression in the Astragalus-treated group was decreased as compared with the diabetes group (P〈0. 01). Conclusion: Tie-2 may play an important role in the pathogenesis of the early stage diabetic renal injury. The reno-protection effect of Astragalus may be mediated by down-regulating the expression of Tie-2 in the kidney tissue of diabetic rats.
