摘 要:目的建立人脐动脉血管平滑肌细胞的体外培养方法。方法运用植块贴壁法进行人脐动脉血管平滑肌细胞的体外原代和传代培养。倒置相差显微镜和免疫荧光染色方法对培养细胞进行鉴定。结果原代和传代培养的细胞生长良好,光镜下细胞呈长梭形"峰-谷"样生长,具有典型的平滑肌细胞特征。经传代培养至第5代,细胞生长特性未见异常改变;经考马斯亮兰R250及α-平滑肌肌动蛋白(α-SMA)免疫荧光染色检测,证实为平滑肌细胞。结论植块贴壁法可成功进行人脐动脉血管平滑肌细胞的体外培养,传代后细胞生物学特征稳定,为慢性肾病并发心血管疾病的机制研究奠定了实验基础。Objective To establish a steady cell model in vitro for vascular smooth muscle cells from umbilical artery. Methods The explant attached method was applied for the in vitro primary culture and subculture of umbilical artery smooth muscle cells. The cultured cells were observed and identified by inverted microscopy and immunofluorescence method. Results The umbilical artery smooth muscle cells primarily cultured and subcuhured grew well with long spindle shape and exhibited typical "hill and valley" pattern under light microscope, with a typical characteristic of smooth muscle cells. The umbilical artery vascular smooth muscle cells had no significant change of biological characteristics after 5 generations of culture. These cells were identified by Coomassie brilliant blue R250 and immunofluorescence method (α-SMA). Conclusion The umbilical artery vascular smooth muscle cells can be successfully cultured by explant attached method, and the cells cultured may have a stable biological characteristics, which lays a foundation for the mechanism study of chronic kidney disease complicated with cardiovascular disease.
