摘  要:目的探讨吡格列酮对血管紧张素Ⅱ(AngⅡ)诱导的血管外膜成纤维细胞(AF)增殖以及Ⅰ型胶原表达的调节作用。方法采用细胞贴壁法培养SD大鼠胸主动脉AF;噻唑蓝(MTT)比色法、流式细胞仪检测吡格列酮对AF增殖及细胞周期的影响。蛋白免疫印迹检测吡格列酮对Ⅰ型胶原表达的调节作用。结果①吡格列酮可抑制AngⅡ诱导的AF增殖,呈剂量依赖性,浓度为10×10-6mol/L时作用最明显(P<0.01);②与AngⅡ组比较,吡格列酮可增加AFG0/G1期细胞,减少S期细胞,并同时降低细胞增殖指数(P<0.01);③吡格列酮可明显抑制AngⅡ诱导的Ⅰ型胶原表达(P<0.05)。结论吡格列酮可能通过抑制AF的增殖和Ⅰ型胶原表达发挥其抗血管重构作用。 Objective To investigate the effects of pioglitazone on angiotensin Ⅱ ( Ang Ⅱ ) -induced proliferation and collagen type Ⅰ expression of adventitial fibroblasts (AF). Methods The AFs were isolated from rat thoracic aorta. MTT colorimetry and flow cytome- try were used to study the effects of pioglitazone on proliferation and cell cycles of AF. The expression of collagen type Ⅰ regulated by pioglitazone was examined by the method of Western blot. Results Pioglitazone inhibited the proliferation of AF in a dose-dependent manner, and the most marked effect could be observed at the concentration of 10 × 10^-6 mol/L for pioglitazone (P 〈0.01 ). Pioglitazone increased the proportion of G0/G1 phase events, decreased the proportion of S phase events and the cell proliferation index( P 〈0.01 ). Pioglitazone attenuated Ang Ⅱ -induced collgen type Ⅰ expression in AF ( P 〈 0.05 ). Conclusion Pioglitazone can inhibit proliferation and collagen type Ⅰ expression of AF and thus plays an important role in improving vascular remodeling related to adventitia.
