摘 要:目的:检测白细胞介素(IL)-18、-6在糖尿病大鼠肾脏的表达,探讨其在糖尿病肾脏病变过程中的作用。方法:40只雄性Wistar大鼠,随机分为正常对照4周(NC1组)及8周(NC2组),糖尿病4周(DM1组)及8周(DM2组),每组10只。DM组按60 mg/kg一次性腹腔内注射链脲佐菌素建立糖尿病大鼠模型。检测各组大鼠体重(BW)、肾重、尿量(UV)及尿微量白蛋白排泄率(UAER)。用免疫组化方法检测各组肾组织IL-18、IL-6的表达,利用计算机图像分析系统进行定量分析。结果:DM组大鼠的肾重指数(KWI)、UV、UAER较NC组大鼠显著增高(P<0.01),DM2组较DM1组增高明显(P<0.05)。IL-18、IL-6在NC组大鼠肾组织中仅有少量表达,DM1组表达较NC组明显增多(P<0.01),DM2组表达增高更为显著(P<0.01)。肾脏局部IL-18、IL-6与UAER呈显著正相关(r=0.471,P<0.01,r=0.513,P<0.01)。结论:炎性反应在糖尿病肾脏局部损害中发挥重要的作用。Objective: To investigate the potential role of IL-18 and IL-6 expression in the renal of diabetic rats. Methods- Forty male Wistar rats were divided at random and equally into normal control group of 4 weeks (NC1 group) and 8 weeks (NC2 group), diabetic mellitus group of 4 weeks (DM1 group) and 8 weeks (DM2 group). The rat models in DM group were induced by injection of streptozotocin (STZ, 60 mg/kg, intra-peritoneally). The expression of IL-18 and IL-6 in the renal tissue was detected by immunohistochemistry staining and computer image-pattern analysis system. At the same time, kidney weight index(KWI), urine volume(UV) and UAER were measured. Results: The levels of KWI, UV and UAER were significantly higher in DM groups (all P〈0.01). The expression of IL-18 and IL-6 was weaker in NC group, while stronger in DM1 group (P〈0.01) and much stronger in DM2 group (P〈0.01). There was a significant positive correlation between the expression of IL-18/IL-6 and UAER (r=0. 462, P〈0.01 and r =0. 513, P〈0.01, respectively). Conclusion: Inflammation factors play an important role in the progression of renal lesion in diabetic rats.
