摘  要:目的探讨噻奈普汀对慢性应激抑郁症模型大鼠海马钙/钙调素依赖性蛋白激酶Ⅱ(α-CaMPKⅡ)表达的影响。方法采用慢性综合应激制作动物模型,同时给予药物干预,运用western blot法观察大鼠海马α-CaMPKⅡ的表达变化。结果应激后抑郁症组体质量(167.30±11.10)g、水平穿越格数(12.88±6.51)格、竖立次数(5.88±3.23)次、修饰次数(7.13±4.02)次,均显著低于对照组[分别为(273.20±15.3)g、(52.00±12.17)g、(23.70±6.95)g、(11.00±1.94)g;均P〈0.05]。应激后噻奈普汀组大鼠体质量(173.60±20.40)g与抑郁症组无明显差异(P〉0.05),水平穿越格数(24.00±9.48)格、竖立次数(14.50±3.95)次、修饰次数(10.80±2.04)次均高于抑郁症组(P〈0.05),但低于对照组(P〈0.05)。抑郁症组大鼠海马α-CaMPKⅡ表达明显减少(P〈0.01),噻奈普汀组与对照组差异无显著性(P〉0.05)。结论慢性应激时α-CaMPKⅡ表达减少,噻奈普汀通过α-CaMPKⅡ途径发挥神经元保护作用。Objective To observe the effects of tianeptine on the expression of α-CaMPK Ⅱ in the rat hippocampus after chronic stress. Methods Chronic stress rats were treated with tianeptine, then western blot was used to observe the changes of α-CaMPK Ⅱ expression in the hippocampus of rats. Results After the chronic stress, the body weight ( 167.30 ± 11.10) g, the horizontal crossing numbers ( 12.88 ± 6.51 ), the erection times (5.88 ±3.23) ,the modification times(7.13 ±4.02) of the depression group are less than control group[ (273.2 ± 15.3)g,(52.0 ±12.17)g, (23.70 ±6.95)g,(11.0 ± 1.94) g; P〈0. 05 ]. There is no obvious difference of body weight between group of tianeptine group and depression group ( P 〉0. 05 ). As for the horizontal crossing numbers ( 24.00 ± 9.48 ), the erection times ( 14.50 ± 3.95 ), the modification times ( 10.80 ± 2.04), they are all more than the depression group( P〈0. 05) ,but less than the control group( P 〈 0. 05 ). Compared to the control group, the α-CaMPK Ⅱ expression of depression group decreased significantly( P〈0.01 ) ,and the α-CaMPK Ⅱ expression of tianeptine group had no difference( P 〉0.05 ). Conclusion The expression of α-CaMPK Ⅱ in hippocampus of chronic stress depression rat decreased. Tianeptine protects neuron by the way of α-CaMPK Ⅱ.
