摘 要:目的探讨抑郁模型大鼠心肌组织中Bcl-xLmRNA和BaxmRNA基因表达的变化及抗抑郁药文拉法辛对其的影响。方法选择48只SD大鼠,随机分为3组,即对照组,抑郁组,药物组,每组16只。采用慢性轻度不可预见性的应激结合孤养制备抑郁模型。抑郁组及药物组大鼠共接受21d各种不同的应激。刺激同时药物组灌胃给药,抑郁组蒸馏水灌胃。采用敞箱实验和糖水消耗实验观察大鼠行为改变。用逆转录聚合酶链反应(RT-PCR)检测心肌组织凋亡相关基因BaxmRNA和Bcl-xLmRNA表达情况。结果应激后,抑郁组敞箱实验得分和糖水消耗量[分别为(34.5±14.9)分、(9.8±4.6)分、(5.6±1.7)g]与对照组[分别为(69.1±21.3)分、(19.2±5.4)分、(9.4±1.4)g]比较明显降低(P〈0.01);与抑郁组比较,文拉法辛能增加抑郁大鼠敞箱实验得分和糖水消耗量[分别为(68.9±17.6)分、(17.8±4.7)分、(8.7±1.2)g,P〈0.01]。抑郁组与对照组比较,抑郁组心肌组织BaxmRNA基因表达明显升高(P〈0.05),Bcl-xLmRNA基因表达明显降低(P〈0.01);与抑郁组比较,文拉法辛可降低抑郁模型大鼠心肌组织BaxmRNA基因表达,升高Bcl-xLmRNA基因表达(P〈0.05)。结论抑郁模型大鼠心肌组织BaxmRNA基因表达升高,Bcl-xLmRNA基因表达降低,而文拉法辛不仅可显著改善抑郁模型大鼠的异常行为,并对心肌组织Bax mRNA和Bcl-xLmRNA基因表达具有干预作用。Objective To study the change of the expression of mRNA of Bcl-xL and Bax in myocardium of depression model rats and the effect of venlafaxine. Methods 48 adult Sprague-Dawley rats were selected and randomly divided into three groups: control group, depression group and drug group with 16 rats in each group. Chronic mild unpredictable stress and separation were used in the depression group and drug group and gastric perfusion was given to them for 21 days. The behavior of rats was detected by open field test and sucrose consumption test. The expression of bcl-xL mRNA and bax mRNA in ratsinyocardium were measured by reverse transcriptionpolymerase chain reaction(RT-PCR). Results After the chronic stress ,compared with the control group rats, the depression group rats showed obvious reduction of squares crossed and rears in open field test and sucrose consumption ( P〈 0.01 ). Compared with the depression group, the squares crossed and rears in open field test and sucrose consumption were increased in the drug group( P〈0.01 ). Compared with the control group, the expression of bcl-xL mRNA was decreased and the expression of bax mRNA was increased in myocardium in the depression group( P 〈 0.05). As compared with the depression group, the expression of bcl-xL mRNA was increased and the expression of bax mRNA was decreased in myocardium in the drug group ( P 〈 0.05 ). Conclusion The expression of bax mRNA was increased and bcl-xL mRNA was decreased in myocardium of the depression model rats, whereas venlafaxine can prevent the abnormal behavior and has antagonism against the expression of mRNA of Bcl- xL and Bax.
