摘 要:目的通过观察醒脑益智方拆方对AD模型大鼠学习记忆及胆碱能系统的影响,以探讨醒脑益智方对AD作用的有效成分。方法15月龄Wistar大鼠100只,随机分成10组:正常组,假手术组,模型组,哈伯因组,醒脑益智方原方组、补肾组、化痰组、活血组、补肾化痰组和补肾活血组。大鼠双侧基底前脑注射IBO和Aβ25-35混合物造成AD模型。动物经中药干预28d后,各组大鼠进行Morris水迷宫行为学试验并检测大鼠海马胆碱能系统的影响。结果本实验结果表明,IBO和Aβ25-35注射所致AD痴呆动物模型组学习记忆能力明显低于正常组和假手术组,表明此AD模型制作成功。在定位航行中,潜伏期醒脑益智方原方[(72.8±9.5)s]和补肾化痰方组[(78.7±8.2)s]比模型组[(105.1±14.3)s]明显缩短(P〈0.05),在空间搜索中,跨过平台次数醒脑益智方原方[(3.46±0.62)次]和补肾化痰方组[(3.67±0.54)次]比模型组[(0.92±0.15)次]明显增加(P〈0.05)。醒脑益智方原方和补肾化痰方之间无显著差异(P〉0.05)。Ach和ChAT活性醒脑益智方原方和补肾化痰方组比模型组明显升高(P〈0.05),2组之间比较无差异(P〉0.05)。结论醒脑益智方各拆方均能在一定程度上改善AD模型大鼠行为学习水平,改善胆碱能系统损伤。但以醒脑益智方原方和补肾化痰方效果最佳,补肾化痰药是其最佳成分。Objective To observe the effects of XINNAOYIZHI decoction and other compatibilities on learning memory and the activity of acetylcholine (Ach) ,choline acetyhransferase (CHAT) and acetylcholinesterase (AchE) in brain and the relation of its two explore the compatibilities of action of XNYZ. Methods 15- month Wistar rats were randomly divided into normal group, sham operated group, AD model group, Huperzine A treated group, XNYZ group, invigorate the kidney group, dissipate phlegm group, promoting blood flow group, invigorate the kidney admix dissipate phlegm group, and invigorate the kidney admix promoting blood flow group, then induced by injection of basal nucleus of Meynert (BNM)with the composition of ibotenic (IBO) and Aβ25-35 to make AD model. After treating for four weeks, Morris water maze was used to assay the ability of space learning memory and radioimmunoassay was used to assay the activity of Ach, ChAT and AchE in brain. Results In the behavior experiment, average incubation period of the XNYZ group(72.8 ± 9.5 s) and invigorate the kidney admix dissipate phlegm group(78.7 ±8.2 s)have obvious shorten compared to model group( 105.1 ± 14.3 s) ( P 〈 0.05 ). In space research , the frequency in span of platform localization of the XNYZ group(3.46 ± 0.62 times) and invigorate the kidney admix dissipate phlegm group( 3.67 ± 0.54times)have obvious increased compared to model group (0.92 ± 0.15 times) ( P 〈 0.05 ) Compared with model groups ( 8.34 ± 1.52,169.33 ± 55.07 ). the XNYZ group( 18.75 ±2.08,330.37 ± 43.28 ) and invigorate the kidney admix dissipate phlegm group( 18.29 ±2.33,317.43 ±48.92) could increase the content of Aeh and ChAT in the hippocampus of AD model rats ( P〈 0.05 ). There is no difference between group and group ( P 〉 0.05 ). Conclusion XNYZ and other eompatibilities possesses a certain improvement of the space learning memory level and repair the function of cholinergic system for AD model rats,
