摘 要:采用改进的UREA法和CTAB法,分别提取4株白粉寄生孢(Ampelomyces quisqualisCes,AQ)的基因组DNA,并运用ISSR标记技术初步研究了AQ的遗传多态性。结果表明,两种方法均能较好地提取到所有供试菌株的基因组DNA,但改进的UREA法较CTAB法更为简单快速,所提DNA用于后续的ISSR扩增试验更能反映AQ种群内部存在着较高的遗传多样性。Two improved methods, modified UREA and modified CTAB were applied to extract the genomic DNA from four isolates of Ampelomyces qusiqualis (AQ) , and the extracted DNA were assessed using ISSR mark to study the genetic polymorphism of AQ. Results showed that DNA of all the supplied AQ isolates were efficiently extracted by two improved methods, furthermore, the modified UREA was a more convenient method and higher genetic polymorphism among AQ isolates could be sufficiently expressed by ISSR amplification.
