鸡传染性支气管炎腺胃病变型分离株H_(95)免疫原基因的克隆与鉴定

摘　　要：用反转录聚合酶链反应（ＲＴ－ＰＣＲ），从纯化的腺胃病变型鸡传染性支气管炎分离株病毒基因组ＲＮＡ扩增出约１．７ｋｂ的ＩＢＶＳ１基因。将该扩增产物于ＨｉｎｄⅢ和ＢａｍＨⅠ位点克隆到ｐＵＣ１８质粒载体中，对重组质粒载体进行酶切分析，得到与Ｓ１基因预期大小一致的插入片断。经Ｓ１探针Ｓｏｕｔｈｅｒｎ杂交检测，表明插入片断为Ｓ１基因。利用该法筛选到４株阳性重组质粒转化子。Using viral RNA extracted from purified IBV virant H 95 isolated from infections proventriculus disease as template, S 1 Glycoprotein Gene about 1.7kb in length have leen amplified using RT PCR. Cloed this PCR produsts into pUC18 in the restriction endonucleiase site of HindⅢ and BamHⅠ, a series recombinant plasmid was acquired. Cleaving these recombinant plasmid with HindⅢ and BamHⅠ, and identified the insert using southern blot with S 1 Gene Probe, four positive recombinant plasmid with S 1 Gene were selected from candidate recombiant plasmid.
