摘 要:目的探讨加味桃核承气汤及其拆方对糖尿病损伤人脐静脉内皮细胞(HUVEC)培养液中内皮素-1(ET-1)、一氧化氮(NO)、细胞间黏附分子-1(ICAM-1)含量影响。方法采用血清药理学方法,制备中药含药血清并添加至体外培养HUVEC中,测定培养液中ET-1、NO、ICAM-1含量,以罗格列酮为对照药物。结果加味桃核承气汤及其拆方(除益气养阴组外)均可降低模型细胞培养液ET-1含量(P<0.05)、升高NO含量(P<0.01),活血组ET-1含量最低,加味方高剂量组NO含量最高;模型组内皮细胞分泌的ICAM-1显著增多,活血组、泻热通下组、原方组、罗格列酮组均可降低ICAM-1含量,与模型组比较有显著性差异(P<0.05)。结论加味桃核承气汤能显著升高体外HUVEC培养液中NO含量,降低ET-1含量,可能对糖尿病血管内皮功能障碍有较好的改善作用,其有效部位可能主要存在于活血组及加味方高剂量组之中,而降低ICAM-1作用则可能存在于除益气养阴组外各拆方中。Objective To investigate the influence of Jiaweitaohechengqi Decoction and its modified formulas on contents of endothilin-1 (ET-1) , nitric oxide (NO) and intercellular adhesion molecules-1 (ICAM-1) in the cultural solution of injured human umbilical vein endothelioid cells (HUVEC) induced by diabetes. Method The serum pharmacological method was applied to prepare Jiaweitaohechengqi Decoction serum and then the serum was added to HUVEC cultured in vitro. The contents of ET-1, NO and ICAM-1 in the cultural solution were detected taking Rosiglitazone Tablets as the control medication. Result Jiaweitaohechengqi Decoction and its modified formulas (except of Yiqiyangyin Formula group) all decreased the contents of ET-1 (P 〈 0.05 ), and increased NO content ( P 〈 0.01 ) in the cultural solution of model cells. The content of ET-1 was the lowest in Huoxue Formula group and NO content was the highest in the high-dose Jiaweitaohechengqi Decoction group. In the model group the ICAM-1 content was increased obviously and decreased in the Huoxue Formula group, Xieretongxia Formula group, Jiaweitaohechengqi Decoction group and Rosiglitazone Tablets group with a significant difference compared with those of the model group (P 〈 0.05 ). Conclusion Jiaweitaohechengqi Decoction can markedly increase NO content and decrease ET-1 content in the cultural solution of HUVEC in vitro. It may relieve the disorder of vascular endothelioid cells induced by diabetes. Its effective fraction maybe presented in Huoxue Formula group and high-dose Jiaweitaohechengqi Decoction group, and its effect of decreasing ICAM-1 maybe presented in all groups except of Yiqiyangyin Formula group.
