摘 要:【目的】获得肉鸡生长抑素2型受体(SSTR2)基因序列并进行序列分析;揭示不同基因型肉鸡结直肠SSTR2 mRNA表达的发育性变化并进行品种间比较。【方法】运用RT-PCR和RACE方法进行肉鸡SSTR2序列扩增相对定量RT-PCR方法研究AA肉鸡和岭南黄肉鸡结直肠SSTR2 mRNA表达的发育规律。【结果】肉鸡SSTR2从起始密码子至Poly(A)长度为2311bp,其ORF与电子克隆序列完全相同,核苷酸序列与人SSTR2的同源性为81.0%,与大鼠SSTR2同源性为79.5%。其翻译的蛋白由371个氨基酸组成,比人和大鼠的SSTR2多2个氨基酸,鸡SSTR2与人的同源性87.1%,与大鼠的同源性为86.3%。AA肉鸡30d的SSTR2 mRNA表达丰度显著高于其他日龄(P<0.05);2d、16d、44d和58d差异不显著(P>0.05)。黄羽肉鸡的16d的SSTR2 mRNA表达丰度显著高于30d、44d和58d(P<0.05),2d、30d、44d和58d之间差异不显著(P>0.05)。不同基因型SSTR2 mRNA表达丰度比较显示,AA肉鸡30d、44d和58d均显著高于黄羽肉鸡(P<0.05)。【结论】不同物种间SSTR2具有较高的同源性;不同基因型肉鸡结直肠SSTR2 mRNA表达的发育模式不同,AA肉鸡生长后期SSTR2 mRNA表达丰度显著高于黄羽肉鸡。[Objective] This study was carricd out to obtain and analyze sequence of somatostatin receptor subtype 2 (SSTR2), and to reveal and compare the ontogenetic expression of SSTR2 mRNA in colorectum of different broiler chicken genotype. [Methods ] The sequence amplification and 3' RACE of chicken SSTR2 was conducted by reverse transcription PCR (RT-PCR) and RACE; The ontogenetic expression of SSTR2 mRNA in colorectum of Arbor Acer (AA) chicken and yellow-feathered chicken was studied by relative quantitative RT-PCR. [Results] The length of chicken SSTR2 is 2 311bp from ATG to Poly (A) and the nucleotide sequence in ORF of AA chicken was completely same with that of an electric clone. The sequence homology were 81.0% and 79.5% identical from that of human and rat, respectively. The protein translated by the nucleotide sequence in the ORF was composed of 371 amino acids while the protein of human and rat were composed of 369 amino acids. Comparison of the chicken SSTR2 amino acid sequence revealed 87.1% and 86.3% identity with human and rat, respectively. The SSTR2 mRNA expression abundance of AA chicken on 30 d was significantly higher than that on other time points (P〈0.05) while the expression abundance had no significant difference on 2 d, 16 d, 44 d and 58 d (P〉0.05). The SSTR2 mRNA expression level of yellow-feathered chicken on 16 d was significantly higher than that on 30 d, 44 d and 58 d (P〈0.05) and there was no difference between 2 d, 30 d, 44 d and 58 d (P〉0.05). SSTR2 mRNA abundance of AA chicken was significantly higher than that of yellow-feathered chicken on 30 d, 44 d and 58 d (P〈0.05). [Conclusion] These results indicated that the homology of SSTR2 was high between species. AA chicken has the different ontogenetic expression of SSTR2 mRNA from the yellow-feathered chicken in colerectum and the expression abundance of AA chicken is significantly higher than that of yellow-feathered chicken during the anaphase of growth.
