摘  要:目的:观察内质网分子伴侣GRP94在瘢痕疙瘩和正常皮肤组织中的表达,探讨GRP94在瘢痕疙瘩发生中的作用。方法:用免疫组织化学法和免疫印迹法分别检测12例瘢痕疙瘩与12例正常皮肤组织中GRP94的表达。结果:①GRP94在瘢痕疙瘩和正常皮肤组织的角质形成细胞、成纤维细胞、炎症细胞、血管内皮细胞以及毛囊、汗腺、皮脂腺等细胞质及少数细胞的核中表达。②表皮中GRP94的表达量在两种组织无显著性差异(P>0.05);真皮中GRP94阳性成纤维细胞数在瘢痕疙瘩组明显高于(1283.33±565.10/mm2)正常皮肤组(238.33±255.62/mm2),(P<0.05)。免疫印迹法实验结果进一步证实了GRP94的含量在瘢痕疙瘩组(0.50±0.07)高于正常皮肤组(0.15±0.03),(P<0.05)。结论:GRP94在瘢痕疙瘩成纤维细胞中高表达,提示内质网应激与瘢痕疙瘩的过度增殖有关。Objective To investigate the relationship between GRP94 and keloid by detecting the expression of GRP94 protein in keloid and normal skin tissue. Methods Immunohistochemistry staining and western blotting were used in 12 cases of keloid and 12 cases of normal skin tissue. Results (1)GRP94 mainly expressed in cytoplasm,partially anchored in nucleus of keratinocytes,fibroblasts,inflamatary cells and endothelial cells.Hair follicles,sweat glands and sebaceous glands were also stained positively. (2)Although the expression level of GRP94 between the two groups in epidermis was no significant difference (P〉0.05). The positively stained fibroblast cell number in dermis was found significantly increased in keloid group (1283.33±65.10/mm^2), compared with normal skin group (238.33±55.62/mm^2) (P〈0.05) .This result was verified by the statistical data from western blotting test.The protein expression level of GRP94 was (0.50± 0.07)in keloid and (0.15±0.03)in normal skin tissue (P〈0.05). Conclusion The overexpression of GRP94 in fibroblast of keloid supports a possible role of endoplasmic reticulum stress in the excessive hyperplasia of keloid.
