摘  要:目的以AngⅡ刺激乳鼠心脏成纤维细胞(CFC),观察CFC的增殖情况以及第10号染色体同源丢失性磷酸酶张力蛋白基因(PTEN)及转化生长因子-β1(TGF-β1)基因表达。方法以AngⅡ(1×10-7mmol/L)刺激分离培养的CFC,以MTT法检测成纤维细胞的增殖情况,以RT-PCR的方法检测PTEN及TGF-β1基因表达。结果MTT测定的结果:32 h后AngⅡ刺激组的吸光值(A)显著高于该时间点对照组(P<0.05),与正常对照组相比,AngⅡ作用组PTEN基因表达显著下降(P<0.05),TGF-β1基因显著上调(P<0.05)。结论在AngⅡ的刺激下,CFC的TGF-β1表达上调,抑制PTEN基因表达,并引起细胞增殖。AIM To observe the effect of proliferation of cardiac fibroblast cells and expression of PTEN and TGF-β1 gene stimulated by Ang Ⅱ in neonate rats. METHODS Cardiac fibroblast cells (CFCs) were cultured and stimulated by Ang Ⅱ ( 1×10^-7mmol/L). Cell proliferation was measured by MTT and expressions of PTEN and TGF-β1 gene were measured by RT-PCR. RESULTS MTT absorbance (A) of Ang Ⅱ -stimulated fibroblast cells was significantly higher than that of the control group( P 〈 0.05 ) after 32 hours. Expression of PTEN increased and expression of TGF-β1 gene decreased compared with those in the control group(P 〈 0.05 ). CONCLUSION When CFCs in neonate rats are stimulated by Ang Ⅱ, they up-regulate the expression of TGF-β1 gene and down-regulate the expression of PTEN gene, thus promoting the proliferation of the cells.
