摘  要:目的:探讨Wistar大鼠体内低剂量甲胺的慢性分布是否能导致氨基脲敏感型胺氧化酶(SSAO)活性上调而诱导内皮的损伤。方法:甲胺组20只、对照组10只分别给以甲胺(50 mg/kg)、NS(50 mL/kg)持续灌胃6个月后,取胸主动脉免疫组化染色,光镜下观察血管假性血友病因子(vWf)的表达;取升主动脉及心内膜透射电镜下观察内皮细胞的超微结构。检测血浆中SSAO活性、NO及甲醛浓度。结果:甲胺组血浆SSAO活性及甲醛浓度均高于对照组,NO浓度低于对照组,但两组比无统计学意义(P>0.05)。vWf在甲胺组内膜、内膜下层均有较明显表达,对照组只有弱表达。甲胺组内皮细胞超微结构呈不规则及浓缩核,对照组内皮细胞呈现正常细胞超微结构。结论:低剂量甲胺在大鼠体内的慢性分布能够诱导内皮损伤。Objective: To explore whether chronic administration of methylamine of mini-dose in Wistar rots might induce the elevation of semicarbazide-sensitive amine oxidase(SSAO)activity and initiate endothelial injury. Methods: The methylamine group(20)and control group(10)were intragastricly administrated with methylamine(50 mg/kg) and NS(50 mL/kg), respectively. After six months, the thoracic aortas were stained with immunohistochemistry and observed the expression of von Willebrand factor( vWf) under light microscope. The ultrastructure in the ascending aorta and endocardium were observed by transmission electron microscopy. SSAO activity, NO and formaldehyde concentration in the plasma were detected. Results: Although the SSAO activities, formaldehyde concentration were higher and NO concentration was lower in the methylamine group, compared with control group, the difference was not statistically significant. The strong expression of vWf could be observed in endothelium and subendothelium in the methylamine group, however, weak expression in the control group. The endothelial ultrastructure in the methylamine group showed irregular and shrinking nucleus, the control group represented normal endothelial morphology. Conclusion: The chronic distribution of methylamine of mini-dose in rats can induce endothelial injury.
