摘  要:目的制备内脏高敏感大鼠模型,研究5-HT 和 CGRP 在内脏刺激信号传入过程中的作用。观察中药肠炎宁糖浆对内脏高敏感大鼠的作用机制。方法 SD 大鼠30只,随机分为对照组、模型组和治疗组,模型组和治疗组给予鸡卵清白蛋白腹腔注射制备内脏高敏感大鼠,对照组腹腔注射生理盐水。造模2周后模型组和对照组予生理盐水、治疗组予肠炎宁糖浆灌胃4周,进行5-HT 和 CGRP 免疫组化染色并作定量分析。结果造模2周后行内脏敏感性评估,大鼠腹璧撤离反射为3分时所需的注水量,模型组较对照组明显减少(P<0.05)。治疗4周后,模型组较对照组、治疗组明显减少(P<0.05)。免疫组化结果:与对照组、治疗组相比,模型组脊髓后角5-HT 和 CGRP 阳性神经纤维和神经元 PI 值显著增高。结论内脏高敏感大鼠脊髓5-HT 和 CGRP 表达上调,说明5-HT 和 CGRP 在内脏刺激信号的传入过程中起重要作用。肠炎宁糖浆可下调内脏高敏感大鼠脊髓5-HT 和 CGRP。Objective To investigate the expression of 5-hydroxytryptamine and calcitonin gene-related peptide neurons and fibers in the lumbosacral spinal cord of visceral hypersensitivity rats, and the mechanism of chang-yan-ning syrup on visceral hypersensitivity rats. Methods Thirty SD rats were divided randomly into model group, control group and treating group. The model group and treating group were induced by injecting chicken egg albumin into abdominal cavity, while the control group was induced by injecting saline. After 2 weeks, the model group and control group were intragastricly administrated with saline, while the treating group was intragastricly administrated with chang-yan-ning syrup. After 4 weeks, quantitative analysis of 5-HT and CGRP were processed by immunohistochemiscal method and computerized image system. Results Visceral sensitivity evaluation based on abdominal withdraw reaction was made after two weeks of intragastric administration. Compared with control group, the dose of injection water decreased in both model group and treating group (P〈 0.05). After 4 weeks of intragastric administration, the dose of injection water decreased in model group compared with control group and treating group(P〈0.05). The immunohistochemical result: Compared with control group and treating group, the positive index (PI) of 5-HT and CGRP in the lumbosacral spinal cord layer increased in model group (P〈0.05). Conclusion The abnormal function of 5-hydroxytryptamine and calcitonin gene-related peptide neurons and fibers in lumbosacral spinal cord might be one of the mechanisms of visceral hypersensitivity. Chang-yan-ning syrup can significantly decrease 5-HT and CGRP in the dorsal horn of lumbosacral spinal cord in rats. It may be one of the mechanisms that decreasing hypersensitivity in rats.
