摘 要:目的:建立小鼠肿瘤中β-榄香烯的含量分析方法并测定静脉给予β-榄香烯固体脂质纳米粒和乳剂1h后小鼠肿瘤中β-榄香烯的浓度。方法:采用HPLC法,色谱柱为Hypersil BDS C18(4.6mm×200mm,5μm);流动相;乙腈-水(90:10);检测波长为208nm;流速为1mL·min^-1。建立小鼠皮下移植性H22肿瘤模型。结果:β-榄香烯与肿瘤中其他组分能很好分离,在0.4~24.0mg·L^-1“范围内线性关系良好(r=0.9995)。相对回收率大于97%。结论:本方法简单、准确、专属性强,可用于测定实体肿瘤中β-榄香烯的含量。OBJECTIVE An HPLC method was developed for determination of β-elemene in tumor. Concentrations of β-elemene at 1 h after iv administration of solid lipid nanoparticles (SLN) and emulsion to H22-bearing mice were determined respectively. METHODS The detection was carried out on Hypersil BIDS C18 ( 4. 6 mm × 200 mm,5μm) by using acetonitrile-water(90: 10) as the mobile phase with detection at 208 nm and the flow rate was 1.0 mL·min^-1. Tumor-bearing mice were established by injecting H22 cells subcutaneously at the right axillary flank of male KM mice. RESULTS β-elemene could be well separated from other components. The linear range was 0. 4-24. 0 mg·L^-1 (r = 0. 999 5), the average recovery was higher than 97%. CONCLUSION This method is accurate and simple in detecting tumor concentration of β-elemene.
