摘 要:目的:探讨凝血酶敏感蛋白-1(TSP-1)在2型糖尿病心肌病(DCM)发病中的作用。方法:采用高脂高热量饮食诱导出胰岛素抵抗,加小剂量链脲佐菌素注射建立DCM动物模型,12周后采用Masson染色、免疫组织化学染色、实时定量逆转录-聚合酶链反应、Western blotting技术,检测左室心肌胶原含量、TSP-1mRNA和蛋白质表达水平的变化。结果:DCM大鼠左室心肌组织胶原含量明显高于对照组(11.01±3.05vs16.92±3.18,P<0.01),TSP-1mRNA和蛋白表达水平均明显高于对照组(0.0089±0.0034vs0.0141±0.0037,P<0.05;96.38±16.80vs129.98±16.96,P<0.05);TSP-1mRNA水平与血糖、心肌组织胶原含量、LVEDP均呈正相关(r=0.762,P<0.01;r=0.717,P<0.05;r=0.658,P<0.05);与LVSP、-dp/dtmax均呈负相关(r=-0.605,P<0.05;r=-0.694,P<0.05);TSP-1蛋白质表达水平与血糖、心肌组织胶原含量、LVEDP均呈正相关(r=0.735,P<0.01;r=0.750,P<0.01;r=0.716,P<0.05);与LVSP、-dp/dtmax均呈负相关(r=-0.633,P<0.05;r=-0.669,P<0.05)。结论:心肌组织TSP-1高表达在DCM心肌间质纤维化的发生中起着重要的作用。AIM: To investigate the expression and significance of thrombospondin - 1 ( TSP - 1 ) in left ventrieular myocardium of type 2 diabetic eardiomyopathy (DCM). METHODS : The rat model of DCM was established by eating a high -fat diet together with injection of low dose streptozocin (30 mg/kg) intrapertoneally. After 12 weeks, the content of collagen was quantified by Masson staining. The mBNA level of TSP - 1 was determined by quantification real - time lit- PCB, while the protein level of TSP - 1 was analyzed by Western blotting and immunohistochemistry. RESULTS: Compared with the control group, the content of collagen in the DCM group was increased greatly (11.01±3.05 vs 16. 92 ± 3. 18, P 〈0. 01 ). The mRNA and protein expressions of TSP- 1 were significantly higher than those in control group (0.0089±0.0034 vs 0.0141±0.0037, P〈0.05;96.38±16.80 vs 129.98±16.96, P 〈0.05). In DCM group, the mBNA and protein expressions of TSP - 1 showed significantly positive correlations with the levels of fasting blood glucose and collagen (r =0.762, P〈0.01; r=0.717, P〈0.05; r =0.735, P〈0.01; r =0.750, P〈0.01). There was a significantly positive correlation of TSP - 1 mBNA level with LVEDP (r =0. 658, P 〈0. 05). In contrast, there was a significantly negative correlation of TSP - 1 protein with LVSP and - dp/dtmax ( r = - 0. 605, P 〈 0. 05 ; r = - 0. 694,P 〈 0. 05 ). There was a significantly positive correlation of TSP - 1 protein with LVEDP ( r = 0. 716, P 〈 0. 05 ). There was a significantly negative correlation of TSP - 1 protein with LVSP and - dp/dtmax ( r = - 0. 633, P 〈 0. 05 ; r = - 0. 669, P 〈 0. 05 ). CONCLUSION: The increased expression of TSP - 1 may play an important role in the development of myocardial interstitial fibrosis in DCM.
