甘蓝型油菜埃芥二体附加系92I1096附加染色体的遗传分析

摘　　要：应用细胞学方法，对甘蓝型油菜埃塞俄比亚芥二体附加系92I1096（B．napu-carinata，AACC＋BⅡ，2n=40）与甘蓝型油菜品种宁油10号（B．napus L．，AACC，2n=38）的杂种F2群体中附加染色体的遗传进行了分析，并应用分子生物学技术对92I1096、埃塞俄比亚芥品种Doddola（B．carinataA．，BBCC，2n=34）、宁油10号和92I1096X宁油10号的F2群体SRAP标记进行了筛选。细胞学研究结果表明，杂种植株体细胞染色体数为2n=39，F2群体单株间体细胞染色体数有2n=38、2n=39、2n=40共3种类型。观察的91个单株中，体细胞染色体数2n=40的有8个单株，占8．79％；2n=39的有24个单株，占24．37％；2n=38的有59个单株，占64．83％。说明附加染色体能够在后代中以单体或二体的方式传递。筛选出2个与该附加系的附加染色体连锁的特异SRAP标记，即me19～em51 795和me24～em56 350，可用于检测该附加系的附加染色体。The inheritance of the additional chromosomes in F2 population derived from a cross between 92I1096, a disomic addition line （ B. napus-carinata, AACC ＋ B 11,2n = 40 ）, and Ningyou 10 （ B. napus L. , AACC ,2n = 38 ） was carried out by cytogenetic method. Specific marker（s） linking to the addition chromosomes from 92I1096, Doddola （B. carinata A. , Brou, BBCC, 2n = 34） , Ningyou 10 and the F2 population were selected out as well by using SRAP technology. Chromosome number in one somatic cell of F1 hybrid plant of 92I1096 × Ningyou 10 was 2n = 39. And chromosome number in the F2 plants was various which could be grouped into three classes of 2n =38, 2n =39, 2n =40. Among the 91 F2 plants observed, there were 8 plants （8. 79% ） with chromosome number of 2n = 38 and 24 plants （24. 73% ） with 2n = 39 and 59 （64. 83% ） plants with 2n = 40. It indicated that the alien additional chromosomes in 92I1096 could be passed on to its offspring in the manner of monosomic or disomic. Two specific SRAP markers, me19 - me51 795 and me24 - em56 350 ,were identified to be related to the additional chromosomes in 92I1096 which could be used as a molecular marker for screening plants carrying the additional chromosomes in the offspring of 92I1096.
