摘 要:以梭子蟹乳化病的病原菌—溶藻弧菌(Vibrio alginolyticus)为抗原制备兔抗血清,利用异硫氰酸荧光素标记的羊抗兔血清建立了梭子蟹乳化病的间接荧光抗体快速诊断技术,并应用该技术对人工感染梭子蟹、养殖现场梭子蟹及养殖水体进行溶藻弧菌检测.结果表明:该技术检测溶藻弧菌有较高的灵敏度,最低检测量为10~5cfu·mL^(-1).人工感染24h后,就能在梭子蟹鳃和血淋巴中检测到溶藻弧菌.现场检测显示表观有病蟹的阳性检出率为90%,表观健康蟹的阳性检出率为20%,说明间接荧光抗体技术不仅可以用于诊断已感染发病的梭子蟹,而且也可检测出带菌但病症不明显的梭子蟹.Presented in this paper is a method of Indirect of Fluorescent Antibody Technique (IFAT) for rapid diagnosis of pathogenic bacteria of emulsification disease in portunus. The antiserum for Vibrio alginolyticus was obtained by immunizing rabbit against Vibrio alginolyticus, the anti-rabbit IgG goat was marked fluresceinisothiocyanate and used in an IFAT detection, The result show that the lowest Fibrio alginolyticus suspension detection limit in laboratory was 10^5cfi/mL. In artificial infection, Fibrio alginolyticus was found after 24h. On worksite, Fibrio alginolyticus were noted by IFAT to be 90% and 20% for the clinically diseased and healthy Portunus trituberculatus, respectively. The results indicate that not only can the IFAT be applied to diagnose the infected Portunus; but also to detect those slightly infected.
