摘 要:目的:观察黄连总碱(CTB)对铝过负荷致大鼠脑损伤学习记忆功能障碍的保护作用。方法:采用AlCl3(Al3+400mg.kg-1)灌胃大鼠,1次.d-1,5d.wk-1,连续1.5mo或3mo,复制铝过负荷致大鼠脑损伤学习记忆功能障碍的动物模型;将CTB(110、55mg.kg-1)和小檗碱(100mg.kg-1)分别在每次铝给予4h后灌胃大鼠。以大鼠被动回避性学习记忆能力和空间识别能力改变、海马病理形态学改变,以及脑组织SOD活性和MDA含量变化为观察指标。结果:铝过负荷能明显导致大鼠被动回避性学习记忆能力和空间识别能力障碍、海马神经元损伤,使其脑组织MDA含量显著升高、SOD活性显著降低;CTB和小檗碱能明显改善铝过负荷大鼠的学习记忆能力障碍,明显减轻铝过负荷大鼠海马神经细胞的损伤,明显阻遏铝过负荷大鼠海马组织MDA含量的增加和SOD活性的降低,且CTB的作用强于小檗碱,并呈显著剂量依赖性。结论:CTB对铝过负荷致大鼠脑损伤学习记忆功能障碍有明显的保护作用。OBJECTIVE: To study the protective effects of Total base of coptis against brain injury induced by aluminum overload in rats. METHDOS: The brain injury model of rats were established via intragastric administration of aluminum trichloride (A1C13 400 mg·kg^-1) once daily 5d·wk^-1 for 1.5 or 3mo,and Total base of coptis (110mg·kg^-1,55mg·kg^-1) and Berberine(100mg·kg^-1) were intragastrically administered 4h following aluminum administration. The main outcome mea- sures were pathomorphological changes of hippocampi, the changes of learning and memory functions, SOD activities and MDA contents of brain tissue in rats. RESULTS: Aluminum overload could markedly induce impairments of avoidant learning and memory function, spatial learning and memory function, and injury of hippocampal neurons in rats, resulting in a significant increase of MOD content but significant decrease of SOD activities in rats. The administration of Total base of coptis as well as Berberine could significantly prevent mice from the impairment of learning and memory function and hippocampal neuron death, significantly inhibit rising of MDA contents and decreasing of SOD activities in a dose-dependent manner. Total base of coptis had a more powerful potency than Berberine.CONCLUSION: Total base of coptis has an obvious protective effect against cerebral damage induced by aluminum overload in rats.
