摘 要:目的:建立以反相高效液相色谱法测定苦参主要有效成分苦参碱含量的方法。方法:色谱柱为Waters C18(250mm×4.6mm,5μm),流动相为甲醇-乙腈-水-三乙胺(35:20:55:0.02),检测波长为205nm,以苦参碱为外标。结果:苦参碱进样量在0.1024~4.0960μg范围内与峰面积积分值呈良好线性关系(r=0.9999);平均回收率为98.92%,RSD=1.91%(n=6)。结论:本法准确、灵敏、分离度高、重现性好,可用于苦参药材和含苦参成方制剂的质量控制。OBJECTIVE:To establish a RP-HPLC method for the determination of Matrine,the active constituent in Radix Sophorae Flavescentis. METHODS: The determination of samples was performed on Waters C18(250mm x 4.6mm, 5μm) with methanol-acetonitrile-water-triethy lamine (35 : 20 : 55 : 0.02)used as the mobile phase. The detector was operated at 205nm, and matrine was added as the external standard. RESULTS: The linear range of Radix Sophorae Flavescentis was 0.102 4-4.096 0μg( r=0.999 9),and the average recovery of which was 98.92% ,RSD : 1.91% (n = 6). CONCLUSION :The method was proved to be accurate, sensitive, reproducible and high in separation, and it can be use for the quality control of Radix Sophorae Flavescentis and compound preparation that contain Radix Sophorae Flavescentis.
